Objective:To study the effect of cluster needling of scalp acupuncture combined with rehabilitation on neurological function and expression of basic fibroblast growth factor (bFGF) and angiostatin (AS) in rats after focal cerebral ischemia. To explore the mechanism of acupuncture and rehabilitation therapy on ischemic stroke.Methods:90 male Sprague-Dawley rats were randomly divided into sham operation group, model group, acupuncture treatment group, rehabilitation treatment group and acupuncture and rehabilitation treatment group, each group was further divided into 3 days,7 days and 14 days subgroups, each subgroup of 6. Permanent cerebral ischemia rat model was established according to Longa’s method. The model group and the sham operation group accepted no treatment, the acupuncture treatment group was treated by cluster needling of scalp acupuncture, the rehabilitation treatment group received the treadmill training, the acupuncture and rehabilitation treatment group was treated by acupuncture and rehabilitation therapy.3 days, 7 days,14 days after operation, the neural function was evaluated with the modified Neurological Severity Score(mNSS), the expression levels of bFGF and AS proteins were detected by Western blotting.Results:1. Neurological Severity Score(mNSS):At each time point after operation(3d,7d,14d), sham operation group did not appear neurological function defect symptoms, the mNSS score of model group was higher than that of sham operation group, and the difference was significant(P<0.05). Compared with the model group, acupuncture and rehabilitation treatment group and acupuncture treatment group, rehabilitation treatment group mNSS scores were lower, the difference was statistically significant(P<0.05). Compared with the acupuncture treatment group and rehabilitation treatment group, the mNSS score of the acupuncture and rehabilitation treatment group was significantly lower(P<0.05).2. Expression of bFGF protein in the ischemic penumbra:At each time point after operation (3d,7d,14d), compared with the sham operation group, the expression of bFGF protein in model group was increased, and the difference was significant(P<0.05). Compared with model group, the expression of bFGF protein was significantly increased in the acupuncture and rehabilitation treatment group, acupuncture treatment group and rehabilitation treatment group(P<0.05). Compared with acupuncture treatment group and rehabilitation treatment group, the expression of bFGF protein in the acupuncture and rehabilitation treatment group was increased, and the difference was statistically significant(P<0.05).3. Expression of AS protein in the ischemic penumbra:At each time point after operation(3d,7d,14d), compared with the sham operation group, the expression of AS protein in model group was decreased, and the difference was significant(P<0.05). Compared with model group, the expression of AS protein was significantly decreased in the acupuncture and rehabilitation treatment group, acupuncture treatment group and rehabilitation treatment group(P<0.05). Compared with acupuncture treatment group and rehabilitation treatment group, the expression of AS protein in the acupuncture and rehabilitation treatment group was decreased, and the difference was statistically significant(P<0.05).Conclusion:1. Acupuncture and rehabilitation therapy can reduce the Neurological Severity Score(mNSS) in rats with focal cerebral ischemia, promote the recovery of neurological function.2.Acupuncture and rehabilitation therapy can upregulate of bFGF protein expression and down-regulate of AS protein expression, promote angiogenesis.
