Objective:Construction of the pregnant rats with chronic periodontitis (CP) model. Explore the expression level of tumor necrosis factor (TNF) and C-reactive protein (CRP)in pregnant rats with chronic periodontitis in animal models. Advanced real-time fluorescence quantitative polymerase chain reaction (RT-PCR), to investigate the chronic periodontitis and preterm low birth weight children (PLBW)the possible mechanism. Provide a theoretical basis for clinical in the early prediction of pretermdelivery and intervention. Methods:Twenty Wistar female rats were randomlydivided into two groups, control (n=8) and experimental group (n=12). The periodontitis model was established in the experimental group. The females and males in the two groups got together four weeks later. Nineteendays after pregnancy all rats were executed and placenta collected. Thedelivery time and neonatal birth weight were recorded and the pathological changes of periodontal tissue observed. Inspection records periodontal index; observe the pathology of the periodontal tissues of pregnant rats to change; RT-PCR analysis of TNF, CRP expression in the placental tissue of pregnant rats. Resμlts:The animal model of chronic periodontitis was successfμlly established. CP group sμlcus bleeding index (SBI)and attachment loss (AL) levels increased significantly, the CP group of SBI (3.60±0.65)higher than the normal control group (0.52±0.52)in CP group AL (161±22)mm] was significantlyhigher than the normal control group[(4±1)mm], thedifference was statistically significant P<0.05. Experimental groupdelivered30offspring and the control group56offspring. The average number of pups born alive per litter in experimental group (4.1±2.2) was significantly lower than in control group (9.2±2.2), P<0.05. The birth weight of pups in experimental group [(5.01±0.43)g] was significantly lower than in the control group[(5.79±0.53)g], P<0.05. TNF expression levels of the experimental group is1.8times the control group; CRP expression levels of the experimental group is1.9times the control group. thedifference was statistically significant (P=0.001). Conclusion:Combined with comprehensive periodontal examination andhistopathological examination todetermine that the experiment successfμlly established model of pregnant rats with chronic periodontitis. By RT-PCR analysis of the CP group and normal control group, TNF and CRP levels were overexpressed in placental tissues of chronic periodontitis in pregnant rats. CP may be adverse pregnancy through the role of inflammatory factors affect the occurrence anddevelopment.
