Objective: To study the mechanism of brain protective effect of Angong NiuhuangWan on experimental cerebral ischemia, and this study may provide a new experimentalbases on the clinical medication for acute ischemic stroke. Methods: The focal cerebralischemia model of rats were established by thread embolism method and healthy maleSprague-Dawley rats were randomly divided into5groups:sham group,model group,Angong Niuhuang Wan group,western medicine group and integrated Chinese medicineand western medicine group. The impaired neural function was evaluated by Stroke Indexand Neurological Symptom Score. The level of cerebral edema was determined, and thecerebral pathological conformation slides were observed under the light microscope byhematoxylin-eosin coloring, the index of the hemorrheology was checked and the level ofserum interleukin10was detected by enzyme linked immunosorbent assay. Results:Different degrees of movement disorder were observed in all rats except sham group. Thebrain coefficient, The cerebral water content, the whole blood viscosity, plasma viscosity,erythrocyte aggregation index,erythrocyte rigidity index and the level of serum interleukin10were obviously increased except sham group and the deformability index wasapparently lower than that in sham group(P<0.05), model group showed statisticalsignificance compared with other groups (P<0.05).All the treatment groups could not onlydecrease the cerebral coefficient,cerebral water content, the whole blood viscosity, plasmaviscosity, erythrocyte aggregation index and erythrocyte rigidity index but also improvedneurologic deficit and increased the deformability index and the serum IL-10in differentdegrees (P<0.05). The effect of combined Traditional medicine and western medicine wasbetter than other groups (P<0.05). There were no statistical differences between AngongNiuhuang Wan and western medicine (P<0.05). Conclusion: Our studies revealed that Angong Niuhuang Wan improved neurological deficit and increased the level of seruminterleukin10,and decreased brain edema and blood viscosity. Accordingly, it may playan important role in protecting brain from ischemia injury and especially during itcombines with conventional western medicine.
