Objective To set up the methods of extraction, separntion and investigate of in vitro activity of protein from Stellera chamaejasme L. Methods The total protein was deposited by using the salting-out of saturation (NH4)2SO4Separating, different molecular weight of proteins was performed by use of the SephaduxG-150, SeprmdexG-100, SephadexG-50. Furethermore, the isolated proteins from the total protein were identified using the technique of SDS-PAGE gel electrophoresis. The method of agar diiffusion was conducted to test the untibacterial activity of total protein. MTT cell culture was used to test the cell activities of total protein and the six separated proteins. Result Six different molecular weight proteins1-6had been gained in the experiment.The molecular weight of proteins were14.4KDa,35.0KDa,28.0KDa,23,0KDa,10.0KDa,94.0KDa respectively by using SDS-PACE gel clectrophoresis; The study of in vitro activity indicated that the total protein was lack of antibacterial activity but had the function of anti-tumor and the activity of toxicity; the proteins of2,3,4,6had the function of anti-tumor and those of the1,3,4had the toxic effect to normal cells. Conclusion In this paper, the methods can be used to extract and isolate the proteins of Stellera chamaejasme L, and study the biological activity of them.
