| GX50is an ingredient of Zanthoxylum,our previous study found GX-50can inhibitthe activity of transcription factor NF-κB. According to reports NF-κB may play arole in regulating aging and genetic blockade of NF-κB in the skin of chronologicallyaged mice reversed the global gene expression program and tissue characteristics tothose of young mice. Thus we decided to determine if GX-50has antiaging effect onskin. In animal study, the effects of GX50on the skin appearance and collagen densityin mouse skin were examined previous study.In vivo, the effects of GX50on the skin of chronologically aged mice were measured.The collagen density was determined by western blot. In cellular level, we confirmedthe anti-aging effect of GX50by SA-β-Gal staining on the human skin fibroblasts.Secreted malondialdehyde (MDA) concentration and intracellular reactive oxygenspecies (ROS) in fibroblast culture were determined. The irritant and allergenic actionof GX50has been determined by Draize eye test. In vivo, the result showed the skintissue of the GX50treated group had a higher collagen content. In celluler level, theSA-β-Gal staining result showed the GX50treated group is less positive ccompared tothe control group and the GX50pre-treated group is less positive compared to theH2O2treated group. The malondialdehyde (MDA) and the intracellular ROSproduction was both down-regulated by GX50treatment. Comparing to the controlgroup, the MDA and the ROS flourescence intensity was down regulaed30.45%and8.03%. The data of GX-50pre-treated group compared to the H2O2group is61.46% and8.36%respectivly. According to the Draize eye test GX50has no irritant andallergenic action. So as a kind of traditional Chinese medicine, GX-50is novelcosmetics candidate against skin aging. |
