Objective Establish the detection methods of microRNA(miRNA) in tissue/serum of ESCC by Real-time fluorescence quantitative PCR (qRT-PCR). Detect the expression of miRNA in tissue/serum of ESCC by these methods and analyze the expression change of miRNA. Discuss the miRNA’s role in the formation and development of ESCC and its value in ESCC diagnosis, treatment and prognosis.Methods Establish the detection methods of miRNA in tissue/serum of ESCC by qRT-PCR. Detect the expression of miR-29a、miR-12、miR-21、miR-205、miR-206and miR-518b in35cases of ESCC cancer tissues and adjacent tissues. Detect the expression of miR-129、miR-21and miR-205in20cases of ESCC patients paired serums and20cases of normal controls serums.Results The expression of miR-21、miR-205is higher in ESCC caner tissue than in adjacent tissue (P<0.05). The expression of miR-129、miR-518b is lower in ESCC cancer tissue than in adjacent tissue (P<0.05). The expression of miR-205in various pathological stage ESCC has significant differences. Expression of miR-21is higher in ESCC serum than in normal control serum.Conclusion1、In the formation process of ESCC miR-21、miR-205may be high expression while miR-129、miR-518b may be low expression.2、The expression of miR-21in serum and cancer tissue increased significantly. Detection of miR-21can be used as ESCC auxiliary diagnostic index in serum.
