Study On Pharmacodynamics And Acute Toxicity On Wendantang Granule Prescription

The modified Wendangtang granule prescription is composed of8herbal medicines as Ginseng, Shichangpu, Chuanxiong, Poligala and so on. It features nourishing kidney and eliminating plegm.In clinic, it can be used for prevention and treatment of senile dementia.This researching paper makes systematic study on its medical effects on hyperlipidemia and insomnia which both belong to the syndrome of phlegm—dampness. And at the same time, to observe the acute toxicity using KM mouse.Methods:1. In this study, animal model of hyperlipidemia in rats hasbeen established by douched high fat emulsion. Compared with Simvastatin Tablets and the capsule of Xuezhikang, the effects of Wendan decoction onserum(TC, TG, HDL-Cand LDL-c), serumApo(A-I, B)and serum(SOD and MDA)in adult rats with chronic hyperlipidem were observed by blood biochemistry and immunoassay.2. Detect the synergistically sedative and sleeping effects of the recipe and Nembutal sodium in mice by using routine pharmacological methods.3. Acute toxicity experiment:20female and20male KM mice each were divided into two groups, compound capsule group and control group, each had10mice. The medicine groups were fed with mecidine in it speak concentration and volume by gavage, the control groups were fed with equivalent physiological saline.1week after the gavage observed the animals’ behavior, drink and food, diarrhea, death; pelage, urine and stooland their main organs’ appearance.Results:Wendan decoction has reduced the contents of blood TC, TG, LDL-C, ApoB and MDA of hyperlipidemia in rats. It has raised the contents of blood Apoa-I and SOD. And the recipe,combined with Nembutal sodium, could promote sleep and decreas autonomic locomotion of mice. At the same time, acute with toxicity experiment showed that the modified Wendangtang granule prescription has no toxic, a good safety.Conclusion:Wendan decoction effectively regulates the lipid metabolism, prevent and treat the hyperlipidemia-induced diseases. It is applicable to adiposis hepatica, coronary heart disease(CHD), scleratheroma. The research also indicates the treatment for insomnia. At the same time, acute with toxicity experiment showed that the modified Wendangtang granule prescription has no toxic, a good safety.
