Objective:To investigate the relation between single nucleotide polymorphisms(SNP) of ALDH2gene and cerebral infarction susceptibility.Method:Lacunar cerebral infarction patients50cases, atherosclerosis cerebral infarction patients50cases and50healthy persons for control. Whole blood DNA extracted by centrifugal column method, augmented by PCR, the polymorphisms of ALDH2gene G487A site were detected by Gene Chip. At the same time, NO, ONOO-,TC,TG,GLU were detected. General clinical data, dangerous factors, frequency of genotype and allele among the three groups and difference of examined data of each genotype among the patients were compared.Result:1、Genotype frequencies:the GG, GA, AA genotype frequencies were84%,14%,2%;80%,18%,2%;58%,38%,4%among lacunar infarction, atherosclerotic thrombotic cerebral infarction and control group. GG genotype and G allele frequency in lacunar infarction group were higher than the controls(χ2=8.208,P=0.004;χ2=7.292P=0.007).Atherosclerotic thrombotic cerebral infarction were higher than the controls (χ2=5.657, P=0.017;χ2=5.103, P=0.024), too.2、Serum TC,TG,GLU,LDL-C but HDL-C and ApoAl were significantly higher in cerebral infarction group than the controls, difference was statistically significant(F<0.05). Apolipoprotein B100(ApoB100) levels were lower and the difference were not statistically significant (P>0.05). History of smoking, drinking histories, diabetes history, history of hypertension incidence and SBP,DBP with cerebral infarction were higher than the control group, the difference were statistically significant (P<0.05), age and gender were not statistically significant (P>0.05).3、The levels of NO and ONOO-of cerebral infarction were higher than control, the difference is statistically significant (P<0.05)4. SBP、NO、ONOO-were different between GG and GA/AA genotype of among lacunar infarction, whereas Glu among atherosclerotic thrombotic cerebral infarction (P <0.05)Conclusion:1、ALDH2gene GG genotype and G allele may be a risk factor for cerebral infarction.2、ALDH2gene GG genotype may increase the risk of lacunar infarction by affection blood pressure and the NO pathway,rather than by acetaldehyde metabolic pathways3、ALDH2gene GG genotype may increase the risk of atherosclerotic thrombotic cerebral infarction by affection GLU.4、Smoking history, drinking history, diabetes history, and dyslipidemia may be the risk factors for cerebral infarction.
