| Objective To find new molecular marker and clinical diagnosis and treatment for esophageal carcinoma, we detect the expressions of5-lipoxygenase (5-LOX) and EGFR in human esophageal squamous cell carcinoma (ESCC) and explore the relationship between theirs up-regulation and tumor occurrence, development and other biological characteristics.Methods Collecting53cases of ESCC specimens and36cases of non-cancerous tissue from the patients who treated by radical surgery of esophageal carcinoma in the First Affiliated Hospital of Xinxiang Medical University from May.2011to Sep.2011. The expressions of5-LOX and EGFR were examined by reverse trans transcription polymerase chain reaction (RT-PCR) and immunohistochemistry. The results were used for statistical analysis combining with some relevant clinical parameters.Results (1) The relative expression of5-LOX mRNA in ESCC tissue was0.659±0.155which was prominently higher than those in the normal esophageal tissues (0.382±0.108),the difference was statistically significant (P<0.001); the average grey scale values of5-LOX protein was51.853±3.793in the ESCC tissue and37.112±3.404in non-cancerous tissue, the difference was also statistically significant (P<0.001).(2) The relative expression of EGFR mRNA in ESCC tissue was0.582±0.155, which was prominently higher than those in the normal esophageal tissues (0.357±0.108),the difference was statistically significant (P<0.001); the average grey scale values of EGFR protein were47.969±2.846in the ESCC tissue and34.165±3.708in non-cancerous tissue, the difference was also statistically significant (P<0.001).(3) The expressions of5-LOX and EGFR were associated with tumor infiltration, histological grade, lymph node metastasis and the clinical stages(P<0.05), but had no relationship with the tumor size, location of cancer and general classification of tumors.(4)the expressions of5-LOX and EGFR in cancer tissue were positively related (P<0.01).Conclusion The expressions of5-LOX and EGFR are higher in ESCC tissues than non-cancerous and this is related to tumor infiltration, differentiation, lymph node metastasis and the clinical stages. A suggests that its high expression may participate in the cancer occurrence and development, and testing5-LOX and EGFR’s expression level will be helpful to understand the biological behavior and prognosis of ESCC. |
