Objective The expressions of excision repair cross completion1(ERCC1) and ribonucleotide reductase (RRM1) were tested in human esophageal squamous cell carcinoma (ESCC) and the correlation between theirs up-regulation and tumor was explored, such as occurrence, development, depth of invasion and lymph node metastasis, to provide the system supporter for the study of ESCC in molecular biology and offer the theory basis to the esophageal carcinoma patient in clinical diagnosis and prognosis.Methods ERCC1and RRM1expressions in62cases of ESCC specimens and36cases of non-cancerous tissue which lie more than5centimeter on the ESCC tissue were examined by immunohistochemistry and reverse transcription polymerase chain reaction (RT-PCR). The results were used for statistical analysted. All of the specimens were collected from the patients who underwent complete surgical resection of esophageal carcinoma in the First Affiliated Hospital of Xinxiang Medical University from Mar.2012to Aug.2012.Results (1) The average gray scale values of ERCC1protein was47.528±2.158in the ESCC tissue and35.586±2.054in the non-cancerous, the difference was statistically significant (P<0.001); The relative expression of ERCC1mRNA in ESCC tissue was0.544±0.088which was prominently higher than those in the normal esophageal tissues0.310±0.138, the difference was statistically significant(P<0.001).(2) The average gray scale values of RRM1protein was50.858±1.737in the ESCC tissue and44.895±2.069in the non-cancerous, the difference was statistically significant (P<0.001); The relative expression of RRM1mRNA in ESCC tissue was0.503±0.103which was prominently higher than those in the normal esophageal tissues0.401±0.089, the difference was statistically significant P<0.001).(3)The expressions of ERCC1and RRM1were associated with depth of invasion, histological grade, lymph node metastasis and the clinical stages(P<0.001), but had no relationship with the tumor size, location of cancer and general classification of tumors.(4)The expressions of ERCC1and RRM1in cancer tissue were positively related (P<0.001).Conclusion The expression levels of ERCC1and RRM1were significantly higher in ESCC tissues than that in non-cancerous. There were significant correlations between intratumoral expression of ERCC1and RRM1.Also,the expression of ERCC1and RRM1related to depth of invasion, histological grade, lymph node metastasis and the clinical stages. This suggests that the high expression may participate in the cancer occurrence and development, testing the expression level of ERCC1and RRM1will be assessed for learning tailor chemotherapy, biological behavior and prognosis in ESCC.
