Objective:To establish the molecular identification methods of黄精Polygonatum sibiricum Red. and Polygonatum cyrtonema Hua, and systematically do the genetics research about Polygonati Rhizoma of different origin, then provide a theoretical basis for its conservation and utilization.Method:(1)Extracting DNA of Polygonati Rhizoma by using a modified CTAB method and the kid method.(2) Using the ITS2,psbA-trnH sequence as the primer for the DNA samples’PCR amplification.(3) Systematically analysis of sequence samples after sequencing by using the DNAMAN and MEGA.Result:(1) The DNA can be extracted from the fresh medicinal materials of Polygonati Rhizoma by the both methods and be used for PCR amplification.(2) The psbA-trnH sequence used for identification of Polygonati Rhizoma resources variety and germplasm genetic diversity is obtained. (3) The Phylogenetic tree of Polygonati Rhizoma group is obtained.Conclusion:(1) The bands of PCR amplification which use the psbA-trnH sequence as the primer are clear and suitable for sequencing and identification of Polygonati Rhizoma resources variety.(3) By establishing the phylogenetic tree, the two varieties of Polygonati Rhizoma can be distinguished, and their germplasm genetic diversity can be analyzed.
