Objective:To investigate the association between the reaction system of gamma-glutamyltransferase (GGT), biomarkers of oxidative stress and vessel disease inpatients with unstable angina pectoris and normal blood pressure. Methods:Unrelated Han Chinese subjectsin Northeast China were enrolled inthis case-control study, which was performed in the Cardiovascular Center, the FirstHospital of Jilin University, Changchun. Patients were diagnosed by coronaryangiography. We measured biomarkers of oxidative stress, such as GGT activity、GGTrelative activity、LDL-HNE、LDL-MDA、Concentration of protein carbonyls、GSH、T-AOC、H2O2、Anti-hydroxyl radical antioxidant capacity. Statistical analysis wasperformed using the nonparametric Kruskal–Wallis test for comparison of vesseldisease and biomarkers of oxidative stress. Moreover, we analyzed the correlationsbetween GGT activity and levels of other biomarkers of oxidative stress by Spearman’scorrelation. Meanwhile, we ploted3-dimensional (3D) figures (independent variables,GGT activity, GGT relative activity、HNE-LDL、Anti-hydroxyl radical antioxidantcapacity or GSH; dependent variables, levels of other biomarkers of oxidative stress),and analyzed the relation between them by response surface regression (RSR). Finally,we ploted ternary figure to analyze the relation between GGT activity, GGT relativeactivity, GSH, HNE-LDL, Anti-hydroxyl radical antioxidant capacity, and diseaseextent quantified by coronary vessels, and analyzed the relation between them by RSR.Results:GGT relative activity and LDL-HNE levels is significantly associated with thenumber of diseased vessels in patients with normal blood pressure and unstable anginapectoris(P<0.05). GSH and the ability to inhibit the formation of hydroxyl radicalswith the increased number of lesions was significantly decreased (P <0.05). GSH has asignificant contribution to the LDL-MDA level, with elevated GGT activity and GSHlevels were significantly higher (P <0.05) in GGT activity-GSH reaction system. GGT relative activity has a significant contribution to the LDL-MDA level, with increasingof GGT relative activity GSH levels were significantly higher (P <0.05).Withincreasing of GSH concentration, the degree of vascular lesions tended to increasewhen GGT activity in0-50Ua/L、GSH in18-28mg/L in GGT activity-GSH reactionsystem, With the reduction of GSH concentration, the degree of vascular lesions tendedto increase when GGT relative activity in0-8Ua/Um and GSH concentrationin12-18mg/L GGT relative activity in5-8Ua/Um in GGT relative activity-GSHreaction system. Conclusions:GGT relative activity can be used as a marker of the degree of vascular lesions innormotensive unstable angina. LDL-HNE with the increased number of lesions wassignificantly higher; GSH significantly reduced with the increased number of diseasedvessels; ability to inhibit the formation of hydroxyl radicals with the increased numberof lesions was significantly decreased in single, double, triple-vessel disease in patientswith unstable angina pectoris and normal blood pressure, In the GGT activity-GSHreaction system, LDL-MDA level, GSH has a significant contribution. In the relativeactivity of GGT-GSH reaction system, GGT relative activity of LDL-MDA level has asignificant contribution. These two reaction system to select the LDL-HNE andLDL-MDA-modified significant differences, they tend to be associated withMDA-modified LDL.
