Background and objective: Malignant tumor is a kind of disease withmain characteristics of inappropriate cell division and differentiation.Previous studies mainly focused on oncogene and tumor suppressor genes,but in recent years, the central point of studies of malignant tumorgradually turned to the abnormal signal transduction mechanisms. Thefundamental reason of the occurrence of malignant tumor lies in disorderof its signal transduction mechanism, thereby generated various abnormalsignal transduction pathways, among which the relevance studies betweengrowth factors mediated pathway and tumor received broad attention ofscholars. The present studies have shown that tumorigenesis is related tothe abnormal expression of growth factors and its receptors. In recentyears,research about insulin-like growth factors (IGFs) system is gettingmore and more attention. As the main pathways activated receptor,IGF-1R is regarded as the key factor of the IGFs axis which turned it intothe focus of cancer research. Review the past research at home andabroad, it showed that high expression of IGF-1R is related with breast cancer, non-small cell lung cancer, prostate cancer, esophageal cancer,colon cancer, ovarian cancer and so on. IGFBP-3is regarded as the mainbinding protein which regulate the IGF signaling pathways. And in manystudy of malignant tumors, IGFBP-3is considered to have two aspects ofeffect including apoptosis and proliferation, therefore became the focus ofstudy. However, the study of correlation between IGFs and oralsquamous cell carcinoma is very lack at home and abroad. To investigatethe correlation between IGF signaling pathways and oral squamous cellcarcinoma, we chose the IGF-1R and IGFBP-3as the observation indexesto observe the rule of the expression of both factors in oral squamous cellcarcinoma, to further clarify the occurrence,development and metastasisof oral squamous cell carcinoma.Methods: In this experiment, we adopted immunohistochemical method,respectively tested the expression of IGF-1R and IGFBP-3in66cases oforal squamous cell carcinoma and20cases of normal oral mucosa. Weanalyzed the data compared with clinical features of patients usingSPSS17.0statistical software. Chi-square test was used in analyzingsample positive rate, spearman test used in analyzing correlation betweenIGF-1and IGF-1R. P<0.05is used as standard for whether the data isstatistically significant.Results: Expression of IGF-1R in oral squamous cell carcinoma wassignificantly higher than the normal group (P<0.01). Rate of the expression of IGF-1R inⅠ-Ⅱperiod group was lower than that in Ⅲ-Ⅳperiod group (P<0.05), and positively correlated with T stage of tumor(P<0.05). With increase of tumor differentiation degree, the expression ofIGF-1R was decreased (P<0.05). Expression of IGF-1R in the tissueswith lymph node metastasis was significantly higher than the group thatwithout lymph node metastasis (P<0.05). There is no relativity betweenIGF-1R expression and the patient’s age, sex and tumor location (P>0.05).The expression of IGFBP-3in oral squamous cell carcinoma wassignificantly higher than the normal group (P<0.01). The expression rateof IGFBP-3inⅠ-Ⅱ period group was significantly higher than that inⅢ-Ⅳ period group. Expression of IGFBP-3in the tissues with lymphnode metastasis was significantly higher than the group that withoutlymph node metastasis (P<0.05). There is no relation between IGFBP-3expression and degree of tumor’s differentiation, T grades, tumorlocation,patient’s age, sex (P>0.05)., The expression of IGF-1R andIGFBP-3was negatively correlated in oral squamous cell carcinomatissues (P<0.05).Conclusions:1. IGF-1R presented high expression rate in oral squamous cellcarcinoma, with significant correlation with clinical stage, tumor size,degree of differentiation and lymph node metastasis. Suggested thatIGF-1R plays an important role in the tumorigenesis of oral squamous cell carcinoma.2. IGFBP-3presented high expression in oral squamous cell carcinoma,but with the increase of clinical stage, the rate of expression reduced,at the same time, the expression in lymph node metastasis group washigher than the group without lymph node metastasis, suggested thatIGFBP-3may participated in the development and metastasis processof oral squamous cell carcinoma, and maybe involved in the tumorsuppressor mechanisms at the same time.3. IGF-1R and IGFBP-3showed a negative correlation in oral squamouscell carcinoma. Both may has a mutual inhibition effect in thedevelopment of oral squamous cell carcinoma...
