Objective:Through exploring the influences on responses of vascular endothelial growth factor (VEGF), endostatin(ENS), microvascular density (MVD) of ectopic and eutopic endometrium of AM model mices, to illuminate the action mechanism of action of pulinghuayuzhitong prescription in treating adenomyosis from the point of view of angiogenesis related factors.Methods:Healthy and not pregnant females,7Weeks of age, mice of ICR were taken as the research objects. Through the abdominal allogeneic mice pituitary uterine transplant surgery, ICR mice became to an instance of the AM model after surgery of three months. The AM mice were randomly divided into pulinghuayuzhitong prescription high dose group, middle dose group, low dose group, Danefukang unguent group, gestrinone group, model group, blank group, after12weeks of continuous dosing, used HE dyeing to observe the digrees of different groups of endometrial glands and stromal invasion of the myometrium, and used SP immunohistochemical dyeing to detect the responses of ectopic and eutopic endometrial vascular endothelial growth factor (VEGF), endostatin(ENS), and according to the CD31marking of ectopic endometrial vascular endothelial cells, measuring of microvessel density (MVD).Results:(1)Pulinghuayuzhitong prescription high dose group, middle dose group,Danefukang unguent group, gestrinone group can reduce the responses of VEGF in the mice model of ectopic endometrium, in which the high dose group can reduce obviously(P<0.01), comparing the low dose group, the difference is very highly significant (P<0.01). Pulinghuayuzhitong prescription high dose group, middle dose group, low dose group, Danefukang unguent group, gestrinone group can significantly reduce responses of VEGF in mice model of eutopic endometrium (P <0.01). (2) Pulinghuayuzhitong prescription high dose group, middle dose group, low dose group, Danefukang unguent group, gestrinone group can reduce the responses of ENS in the mice model of ectopic endometrium (P<0.01). Pulinghuayuzhitong prescription high dose group, middle dose group, low dose group, Danefukang unguent group, gestrinone group can reduce the responses of ENS in the mice model of eutopic endometrium (P<0.01).(3) Pulinghuayuzhitong prescription high dose group, middle dose group, low dose group,Danefukang unguent group, gestrinone group can reduce the responses of VEGF/ENS in mice model of ectopic endometrium, in which pulinghuayuzhitong prescription high dose group and gestrinone group can reduce obviously (P<0.01). Pulinghuayuzhitong prescription high dose group, middle dose group, Danefukang unguent group can decrease the ratio of VEGF/ENS in the mice model of eutopic endometrium, but no statistical significance (P>0.05).(4)Pulinghuayuzhitong prescription high dose group, middle dose group, low dose group, Danefukang unguent group, gestrinone group can reduce the responses of MVD in the mice model of ectopic endometrium (P<0.01). Pulinghuayuzhitong prescription high dose group, middle dose group, low dose group, Danefukang unguent group, gestrinone group can increase the responses of MVD in the mice model of eutopic endometrium, in which pulinghuayuzhitong prescription high dose group, middle dose group and low dose group can increase obviously (P<0.01), comparing with Danefukang unguent group and gestrinone group, the difference is very highly significant (P<0.01).Conclusions:(1) Pulinghuayuzhitong prescription can reduce the responses of VEGF in the mice model of ectopic and eutopic endometrium, although also lowered the responses of ENS, but AM mice ectopic endometrium VEGF/ENS ratio significantly decreased, and responses of MVD downward trend, which explains pulinghuayuzhitong prescription can restrain ectopic endometrium blood vessel growth through the lower VEGF/ENS ratio. (2) Pulinghuayuzhitong prescription can significantly reduce the responses of VEGF in AM mice model of ectopic and eutopic endometrium, comparing the low dose group, the difference is very highly significant, indicating that the concentration of different drugs have different influence on the responses of VEGF, and drug concentratio is positively correlated with the influence degree.(3) Compare pulinghuayuzhitong prescription high dose group and gestrinone group with pulinghuayuzhitong prescription middle dose group, low dose group and Danefukang unguent group, can significantly reduce VEGF/ENS ratio of AM mice ectopic, indicating that pulinghuayuzhitong prescription high dose group and gestrinone group inhibiting angiogenesis curative effect is significantly better than any other group.(4) Pulinghuayuzhitong prescription reduce menstrual quantity may be due to inhibit angiogenesis of eutopic endometrium and reduce the number of eutopic endometrial vascular.Pulinghuayuzhitong prescription can narrow lesions and decrease the dysmenorrhea degree, which may be due to inhibit angiogenesis of eutopic endometrium and reduce the number of eutopic endometrial vascular.
