Pulsatilla chinensis (Bunge) Regel is an important traditionalChinese medicine used for the treatment of bacterial infection, amoebicdysentery and vaginal trichromoniasis. Pulsatilla chinensis Regel belongsto the genus Pulsatilla (Ranunculaceae), which showed several kinds ofmedicinal effects, such as antitumor, antivirus, antioxidation and so on.Triterpene saponins were the mainly effective constituents from P.chinensis exhibiting activities against cancer, which were reported by YeWen-cai and Yoshihiro.Based on that, In order to find the bioactive compounds and evaluatethe quality of plant materials of P. chinensis from different regions,exploit and utilize this medicinal plant, the studies on chemical constituentsof the roots of Pulsatilla chinensis (Bunge) Regel were carried out.Forty-four compounds were isolated and purified by resin, silica gel, ODSs i l i c a g e l, S e p h a d e x L H-20, m e d i u m p r e s s u r e c o l u m nchromatography,dynamic axial compression chromatography andsemi-preparative liquid chromatography, from70%ethanol extraction of P.chinensis (Bunge) Regel. In addition,the structures of forty-fourcompounds were elucidated by chemical propertyes, and spectroscopicanalysis, mainly1D and2D NMR and MS spectra. In the forty-foursubstances, compounds P4, P5, P6, P7, P8, P15, P16, P21, P22and P24were obtained from this genus for the first time, compounds P11, P26, P33,P36, P38, P40, P42, P43and P44were the previously unknown compounds, and their structures were identified as:Oleanolic acid3-O-α-L-rhamnopyranosyl-(1â†'6)-β-D-glucopyranosyl-(1â†'4)-β-D-glucopyranosyl-(1â†'3)-α-L-rhamnopyranosyl-(1â†'2)-α-L-arabinopyranoside (P11),3-O-β-D-glucopyranosyl-hederagenin23-O-α-D-ribofuranoside(P26),23-hydroxy-3β-[(O-β-D-glucopyranosyl-(1â†'4)-α-L-arabinopyranosyl)oxy]l u p-20(29)-e n-28-o i c a c i d28-O-β-D-glucopyranosyl-(1â†'6)-β-D-glucopyranosyl ester (P33),23-hydroxy-3β-[(O-β-D-glucopyranosyl-(1â†'4)-[α-L-rhamnopyranosyl-(1â†'2)]-α-L-a r a b i n o p y r a n o s y l) o x y] l u p-20(29)-e n-28-o i c a c i d28-O-β-D-g l u c o p y r a n o s y l-(1â†'6)-β-D-glucopyranosyl ester (P36),3β-[(O-β-D-glucopyranosyl-(1â†'3)-α-L-rhamnopyranosyl-(1â†'2)-α-L-arabinopyranosyl)oxy]lup-20(29)-en-28-oica c i d28-O-β-D-glucopyranosyl-(1â†'6)-β-D-glucopyranosyl ester (P38),3β-[(O-β-D-glucopyranosyl-(1â†'3)-α-L-rhamnopyranosyl-(1â†'2)-α-L-arabinopyranosyl) o x y] l u p-20(29)-e n-28-o i c a c i d28-O-α-L-rhamnopyranosyl-(1â†'4)-β-D-glucopyranosyl-(1â†'6)-β-D-glucopyranosyl ester (P40), Pulsatilla triterpenic acid A (P42),Pulsatilla triterpenic acid B (P43) and Pulsatilla triterpenic acid C (P44).On the basis of the chemical studies, plant materials of P. Chinensisfrom nine different regions has been investigated, with8compounds (P1, P2,P9, P18, P20, P23, P34, P39) as the reference substances. This couldprovide a method to control the quality of the medical material. And at thesame time, the saponins’ bioactivities had been tested by MTT assay, and wegot the antitumor structure-activity relationship from the IC50valuecorrelated with its structure. Through the SAR studies, P10was found to be a potential antitumor leading compound.
