Pulsatilla chinensis (Bge.) Regel. belongs to Ranunculaceae, is aperennial herbaceous plant. Potentilla chinensis Ser. belongs to Rosaceae.Although they belong to different kinds of plants, but in actual practice,duo to historical reasons, there are a number of places in china used tocall Potentilla Chinensis Ser. to Anemone, this leads to misusage and maythreaten the recipients health. Therefore, identification Pulsatillachinensis (Bge.) Regel.from Potentilla chinensis Ser. is necessary.There are a lot of traditional methods, such as microscopicidentification, thin-layer chromatography, HPLC and chemicalidentification, etc for identification of Chinese medicine. But there arelimitations in these methods, because these methods are easily affectedby many factors, as a result, the accuracy and reproducibility of thesemethods are not so good, so they are in application to the identificationof medicinal plants. Gene identification is relatively stable, lesssusceptible to temperature, mechanical forces, influences ofenvironmental factors, with good reproducibility and high accuracy. Soit is increasingly applied in plant identification.In this study, we extracted genomic DNA from Pulsatilla chinensis(Bge.) Regel. and Potentilla chinensis Ser, amplified the region of ITSand trnL-F using the universal primer of the angiosperm, and thensequenced the amplified products directly. We compared the two sequencesand found the restrictive site to identify them with BtgI and RsaI. Wedigested and identified the ITS and trnL-F sequences of Pulsatillachinensis (Bge.) Regel.and Potentilla chinensis Ser..These provided anew、 reliable method for identification of Pulsatilla chinensis (Bge.)Regel.from Potentilla chinensis Ser..
