Herba Epimedii belonging Epimedium of Berberidacea is a perennial herb which is widely distributed in the world. Among them59varieties distributed around the world, and China alone had about48varieties, becoming one of the extensive distribution area.5varieties-Epimedium wushanense, Epimedium sagittatum, Epimedium pubescense, Epimedium brevicornum and Epimedium koreanum were collected as quality goods in Chinese pharmacopoeia (edition2010). It is warm, non-toxic and useful to enhance kidney function, strengthen muscles, bones and heart as a crude drug in traditional medicine. The modern pharmacological results indicated that the main active compounds of Epimedium were total flavones and polysaccharides, which could cure many diseases especially applied to heart brain blood vessel diseases and tumors, had wide and remarkable medicine value, developing prospects and had become the hotspot of modern medicine.At present, the research to Epimedium was mostly focused on flavonoids, especially on its pharmacology and medicine effects, but the investment on extraction for active compounds was relatively little. Epimedium Polysaccharides(EPS) had a lot of bioactivity which was superior or different to total flavones, had comprehensive develpping prospect and important position and role. This research used Epimedium acuminatum as experiment materials, optimized a kind of two-step extraction tecnology of total flavones and polysaccharides from Epimedium, aimed to improve extraction efficiency of total flavones and polysaccharides and comprehensive utilization rate of Epimedium under lower energy consumption condition. The antioxidant activity of polysaccharides was evaluated from scavenging rates to3kinds of free radicals, DPPH, O2-and·OH, which could also realize the futher evaluation to the technology. The main results were as follows:1. The two-step extraction technology of total flavones and polysaccharides from Epimedium was optimized:the best conditions of extracting total flavones were that extraction solvent70%(volume fraction) ethanol, ratio of solid to liquid1:10(g/mL). extraction temperature60℃, ultrasonic power150W, extraction time5min and extraction times2; the optimal extraction conditions of polysaccharides were that the residue added distilled water10times volume(g/mL) was extracted under90℃2times, each time lasted30minutes.2. The two-step extraction technology of total flavones and polysaccharides from Epimedium was evaluated:the yield of total flavones having been purified from this technology was7.64%(content of icariin was9.56%), superior to Soxhlet extraction and microwave-assisted extraction method obviously, a little better than previous ultrasonic method, while icariin with no evident difference; the yield of polysaccharides was3.68%, superior to water-boiling and microwave-assisted extraction method.3. Antioxidant activity experiment indicated that EPS had strong capability to scavenge DPPH, O2-and certain capacity to scavenge·OH. Polysaccharides from this optimized technology was inferior to other methods on scavenging DPPH and-OH while nearly same as polysaccharides from water-boiling, microwave-assisted and enzyme-assistant extraction methods on scavenging O2-, except the worst ultrasonic method; in addition, the scavenging capacity to this3radicals of polysaccharides from water-boiling, microwave-assisted extraction method was same too.4. The technology was further evaluated from yield and antioxidant activity of polysaccharides through comparison to other extraction methods:on total efficacy to scavenging DPPH and-OH, polysaccharides from the residue was the least while superior to ultrasonic, water-boiling and microwave-assisted extraction methods on scavenging O2-Though antioxidant function had some losses, the technology of extracting polysaccharides from residue was feasible, could provide the theoretical basis for rational comprehensive utilization and scale industrial production of active compounds of Epimedium.
