Pine pollen, named Song Huang in Chinese traditional pharmacy, has functionsin losing weight and tonifying recorded in ancient medical book. Recently, numerousstudies about pine pollen on lipid metabolism showed that pollen of different speciescan cure or prevent more than60kinds of diseases of human beings. However, thespecific mechanism has not been scientifically understood. To investigate preventingeffects on lipid metabolism of ethanol-extraction from Masson Pine Pollen, in thisstudy4weeks old male-KM white mice (body weight was19+1g) were dividedrandomly into six groups for ten mice at each group: normal control group(NC),positive control group(orlistat, PC), high fat group(FC), ethanol extractlow-dose(EEL), middle-dose(EEM) and high-dose group(EEH)(0.0625g/kg bw d、0.125g/kg bw d and0.25g/kg bw d). Mice were treated by intragastric administrationeveryday for6weeks.Body weight and body fatFC group had significant weight gain (20.50g) compared with NC (P<0.01), thatmeant the establishment of obesity model was successful. Compared with FC group,three ethanol-extract groups decreased obviously (P<0.01) at final weight, weight netincrease and weight gain rate. Body fat content decreased to varying degrees (vs. FCgroup) but did not show significant difference, the rate of body fat is the same too.Blood lipidCompared with FC group, TC level of EEH group had significantly reduced(P<0.05); All three ethanol-extract groups had significantly lower TG level (P<0.01)and higher HDLC level (P<0.01) than FC group.AdipokinesLEP levels of three ethanol-extract groups increased slightly but did not show significant difference. ADP level of EEM group remarkably increased (P<0.01vs. FCgroup). Resistin levels of three ethanol-extract groups had high significant increase(P<0.01vs. FC group)CPT-I content and gene expression of FAS and CPT-I in liverCPT-I contents of three experimental groups were higher than in FC group(P<0.01); But in figure1, EEH was the only group that the gene expression levels ofFAS and CPT-I in liver increased remarkably (P<0.01vs. FC group).Lipids in fecesFeces of each group were collected every6days. TC contents in threeethanol-extract groups increased in different degree compared with FC group (P<0.01)from the second time. TG content in excrement of three ethanol-extract groupsdecreased at different extent compared with FC group.Contents of neuropeptides in blood and gene expression in hypothalamusorexigenic neuropeptide NPY and AgRP levels in blood of three ethanol-extractgroups were significantly lower than FC group (P<0.01). Anorexigenic neuropeptidePOMC decreased as well (P<0.01vs. FC group). On the contrary, anorexigenicneuropeptide CART increased, especially EEM and EEH groups were bothsignificantly higher than FC group (P<0.05). mRNA levels of neuropeptides wereshown in figure4, the trend of changes in gene expressions were opposite to those inblood. NPY expression levels of three ethanol-extract groups were all higher than FCgroup (P<0.01) and AgRP expression of three ethanol-extract groups increased atdifferent extent compared with FC group, POMC expression of EEL group wassignificantly higher than FC group (P<0.05); Similarly, CART of threeethanol-extract groups had significantly declined (P<0.01). Results were opposite tothe changes in blood.In conclusion, masson pine pollen ethanol-extraction could significantly improvelipid metabolism on mice. It proved that masson pine pollen ethanol-extraction has important significance in preventing obesity.
