Benign prostate hypertrophy (BPH) is a common disease of old men, and affectsthe quality of elderly men’s life seriously. At present, the mainstream therapy of BPHwas5α-reductase inhibitors and α1-adrenaline receptor blockers, but they all showedsome side effects. Masson pine pollen extract (MPPE) is a kind of natural plantextracts which with excellent anti-inflammatory and antioxidant activities. It is a plantagents have many obvious advantages such as mild, persistent and stable curativeeffect, less toxicity and adverse reaction. In present study, we replicated theexperimental model of BPH mice by intraperitoneal injection of testosteronepropionate to study the effects of MPPE and SQ, PGY compounds; and to providebasis for clinical treatment of BPH. The experiment included the following sections:1. The preventive and therapeutical effects of BPH in mice: The preventionexperiment lasted6weeks. Mice in three MPPE groups administrated MPPE bygavage for two weeks in advance. Next four weeks, mice administrated testosteronepropionate intraperitoneally every day, and administrated different dose of MPPE inthe same time. At the end of experiment, determinated the physiological indexes andmade paraffin section of prostate tissues to observe the morphological changes. Inaddition, mouse urine was collected for metabonomics analysis.The results displayed that morphological structure of prostate tissue in BPHmodel mice showed obvious BPH characteristics. Marked thickening and hypertrophyas well as hyperplasia with polyps formation in the lining epithelium of the acini wereobserved. Effects of3doses MPPE was certainly different.20mg/kg MPPE effect isa million to one, but concentrations of inflammatory cytokines, DHT and NO in theother two group mice serum decreased significantly. The activity of NOS and PACPwere also declined.80mg/kg MPPE work best. PLS-DA results are basically identical. Completely separation of the control group and model group showed obvious changeshad taken place in the urine metabolites of BPH mice. Finasteride has best effect onthe improvement of BPH. MPPE’ effect is noticeable, but it did not make micerestored to health completely.2. The therapeutical effect of MPPE on BPH in mice: What different withpreventive effect is in the therapeutical experiment, mice firstly administratedtestosterone propionate(i.p.)to induce the BPH disease daily for4weeks, and thenreceived MPPE respectively for4weeks. The experimental results show that prostateweight, prostate index all increased significantly, histomorphology results in prostatetissue showed obvious characteristics of BPH. Effects of3doses MPPE was certainlydifferent.20mg/kg MPPE effect is a million to one, and80mg/kg MPPE work bestThe curative effect of MPPE showed certain dose dependent. PLS-DA results showedobvious changes have taken place in the urine metabolites of BPH mice. Finasteridewas most efficient. MPPE’ effect is noticeable, but it did not make mice restored tohealth completely.3. Effects of MPPE and SQ, PGY compounds on BPH in mice: Specific stepswere in accordance with therapeutical experiment. At the end of experimentdeterminated the physiological indexes. The experimental results show that SQ, PGYcompound showed stronger resistance to BPH than MPPE, and SQ is more efficientthan PGY. But there were no statistical significance.The above results showed that:(1) the preventive effect of MPPE was superior tothe therapeutical one. May be it because MPPE played a protective role timely whenadministrated MPPE while modeling. The chances of suffering from BPH werereduced.(2) SQ, PGY compound showed stronger resistance to BPH than MPPE, andSQ is more efficient than PGY. MPPE compounded with Chinese herbal medicine(SQ and PGY) could be more efficient in improving the BPH disease.(3)MPPE couldsignificantly reduce the pathological symptoms and biochemical parameters of BPHand adjust the level of the significant metabolites. And the mechanism may be relatedto the inhibition of inflammation and oxidation resistance.
