Objective:Substance P(SP) and calcitonin gene-related peptide(CGRP) are the earliest one of the neuropeptides found, which have diverse physiological effects in various tissues and can control the immune response. SP and CGRP containing nerve not only is co-exist in gingival tissue, and affect similarly the neurogenic inflammation induced tooth and periodontal tissue.The bidirectional communication between the immune and nervous systems plays a role in the development of inflammatory gingival tissue. The aim of this research is to detect the differences of the tissue distribution and cellular localization of SP and CGRP in normal and periodontitis gingival tissue and prove nerve systems might contribute to disease development and progression.Thus provide a basis to further illustrate the pathogenesis of periodontitis.Material and Methods:20Gingival tissue samples in health group from health subjects were obtained in extraction of impacted wisdom tooth and supernumerary teeth,20Gingival tissue samples in test group from adult with periodontitis were collected during periodontal surgery for periodontal disease. Power Vision TM two steps immunohistochemical method with anti-SP and anti-CGRP was used to detect the distribution and cellular localization of SP and CGRP in normal and periodontitis gingival tissue in normal and in flammatory gingival tissues between health adult and adult with periodontitis.Results:1. There were few expression of SP in epithelial layer of normal gingival tissue, while, there were positive expression SP in the periodontitis than that in normal gingival tissue, in addition, the cytoplasmic in the spinous cells and inflammation cells of connective tissue had widely claybank-colored,which were heavier colored in inflammatory cells, difference of expressions SP between two groups was significant statistically(p<0.05).2. The expression CGRP was lower in normal gingival tissue, while it was higher and stronger in the periodontitis than that in normal gingival tissue, especially in epithelial layer.There was a significant difference between normal and inflammatory gingival tissues(p<0.01).Conclusion:1. Substance P and CGRP may play an important role in the development and progression of the gingival tissue inflammation.2. Nerve factors may play a role in the pathogenesis of periodontitis and in the reparation of periodontal tissue during periodontitis.
