Objective To investigate the effect of vacuum sealing drainage (VSD) on the NGF expression and the change of the microvascular number in the tissue of chronic wounds, and to study the mechanism of accelerating chronic wounds healing.Methods Ten patients with chronic wounds were as study objects, including1case of soft tissue defect of chest,1case of osteomyelitis with bone exposed and1case of large area skin defect after amputation stump after large area skin defect,2cases of wound infection after skin avulsion injury,2cases of shinbone osteomyelitis,3cases of postoperative wound infection. The skin in the edge tissue and granulation tissue were collected at before the VSD treatment, on the7th days and the14th days after the VSD treatment. The NGF expressions and the microvascular number in the tissues were measured by the immunohistochemical technology. The wound changes were also observed.Results1. The NGF expressions in fibroblasts and endothelial cells on the7th days and the14th days after VSD treatment was significantly higher that than pre-treatment (P<0.001).2. The microvascular number was increased from1.3±1.2of pre-treatment to3.4±1.3of the7th days (P<0.05) and4.9±2.0of the14th days after VSD treatment (P<0.01).3. The VSD treatment reduced the necrotic tissue, purulent secretion and swelling the wounds. Simultaneously, the granulation tissue changed gradually fine granular, bright red, and bleeding easily.Conclusion The VSD promotes the healing of the chronic wounds by increasing the NGF expressions and the angiogenesis in the edge tissues of wounds.
