Objective:In order to explore the material basis of Banxiaxiexin decoction regulating gastrointesti-nal motility, and discuss what mechanism it have.Method:Gastrointestinal motility disorder rat model was established through the way of injection of atropine sulfate subcutaneous and chronic water immersion stress respectively. We screen effective group and the effective parts of Banxiaxiexin decoction regulating gastrointestinal motility by means of measuring remnant rate of stomach and progradation rate of small intestine. Plasma level of gastrin, motilin, somatostatin, vasoactive intestinal polypeptide were measured by utilizing radioimmunoassay; content of c-kit protein in the gastric antrum and jejunum tissues were estimated by utilizing enzyme-linked immunosorbent assay (ELISA) respectively; meanwhile We observe pathology of gastrointestinal tissue and ultrastructure of gastric smooth muscle cells, in order to discuss mechanism of effective group of Banxiaxiexin decoction regulating gastrointestinal dysfunction in rats.Results:Experiments result showed that Banxiaxiexin decoction has a role in facilitating gastric emptying of atropine sulfate-induced gastrointestinal dysfunction in rats, the bitter drugs for purgation also has a role in facilitating gastric emptying of normal rats and atropine sulfate-induced gastrointestinal dysfunction in rats, the pungent drugs for dispersing and the tonic with sweet have no effects on gastric emptying obviously. The ethanol extract of Banxiaxiexin decoction and the bitter drugs for purgation both have a role in facilitating gastric emptying of normal rats and atropine sulfate-induced gastrointestinal dysfunction in rats, but have no effects on progradation of small intestine obviously. With regard to chronic restraint and water immersion stress-induced gastrointestinal dysfunction in rats, the effects on gastric emptying and progradation of small intestine of the bitter drugs for purgation and the ethanol extract of it, which are no significant difference compared with rat model, however, there is a tendency for the ethanol extract of bitter drugs for purgation and itself to facilitating gastric emptying as well as improving progradation of small intestine. With regard to normal rats, atropine sulfate and chronic restraint and water immersion stress-induced gastrointestinal dysfunction in rats, the ethanol precipitation of Banxiaxiexin decoction and the bitter drugs for purgation have no effects on gastric emptying and progradation of small intestine obviously.With regard to chronic restraint and water immersion stress-induced gastrointestinal dysfunction in rats, the ethanol extract of Banxiaxiexin decoction and the bitter drugs for purgation both reduce plasma level of vasoactive intestinal polypeptide, meanwhile increase plasma level of somatostatin, which are significant difference compared with rat model (P≤0.05); the ethanol extract of Banxiaxiexin decoction and the bitter drugs for purgation both increase content of c-kit protein in the gastric antrum, which are significant difference compared with rat model(P≤0.05).Chronic restraint and water immersion stress-induced gastrointestinal dysfunction in rats of which epithelial cells of intestinal villus tips on jejunum mucosal surface exist local focal or extensive degeneration, necrosis, inflammation; gastric mucosal surface exist minor lesion. Jejunum and stomach of rats exist lesser lesion after intragastric administration the ethanol extract of Banxiaxiexin decoction and the bitter drugs for purgation respectively. chronic restraint and water immersion stress-induced gastrointestinal dysfunction in rats, smooth muscle cell nucleus chromatin accumulated, swelling of mitochondrion large area even vacuolation; swelling of mitochondrion relieve after intragastric administration the ethanol extract of Banxiaxiexin decoction and the bitter drugs for purgation.Conclusions:Banxiaxiexin decoction play the role of regulating gastrointestinal motility, the bitter drugs for purgation are the effective group mainly and the ethanol extract parts are the effective components. The mechanism of Banxiaxiexin decoction and the effective group regulating gastrointestinal motility may be relevant to decreasing plasma level of vasoactive intestinal polypeptide, elevating plasma level of somatostatin, increasing content of c-kit protein in the gastric antrum, improving gastrointestinal tissue lesion and mitochondria of smooth muscle cells lesion.
