Background and ObjectivesBladder cancer is the fifth largest human common malignant tumor, its incidence and mortality is in the first place of male urogenital tumor.Bladder urothelial carcinoma (BUC) is the most common, squamous carcinoma of bladder and bladder adenocarcinoma only account for about5%. Squamous carcinoma of bladder and bladder adenocarcinoma are often invasive and metastatic, develop quickly, and have a high malignancy degree.When they are diagnosed they are almost at advanced stage, so the diagnosis and prognosis is not ideal.The HER2gene is a member of the epidermal growth factor receptor family, which encodes cell membrane glycoprotein with tyrosinase activity, mainly expressed in embryonic development, no expression at adult. In recent years, the HER2protein as a prognostic marker of tumor progression is widely concerned. the foreign scholars studied the relationship of HER2gene and bladder cancer.Demonstrated that the HER2protein was expressed in the part of bladder carcinoma, and the clinical stage and pathological grade in these patients is high, the prognosis is poor. At present, the research on HER2in bladder cancer are more concentrated in the BUC, the HER2in bladder squamous cell carcinoma and adenocarcinoma of bladder were rarely reported. In order to investigate the differential expression of HER2in bladder urothelial carcinoma, bladder adenocarcinoma and squamous cell carcinoma, using fresh samples of different histological types and pathological specimens of clinical diagnosis and treatment, we study the expression of HER2protein in different types of tumor tissues by immunohistochemistry and Western blot technique, the relationship between HER2expression and bladder cancer types, pathological grading and clinical staging, we want to discover the usful bladder tumor marker that provides the reference for the clinical diagnosis and treatment.Materials and Methods1:Immunohistochemical method was used to assess the expression of HER2in73cases of bladder cancer specimens (bladder urothelial carcinoma in38cases,18cases of squamous cell carcinoma, adenocarcinoma17cases).These specimens included wax samples and fresh specimen.The fresh specimen were obtained from June2011to October2012and the wax samples were obtained in recent years both from bladder cancer patients hospitalized in the Department of Urology of Second Affiliated Hospital and The first Affiliated Hospital of Zhengzhou University. Exploring the relationship between the expression level of HER2in bladder tumor tissue and different histological type, the clinical staging, pathological grading.2:The expression of HER2in20fresh bladder cancer specimens (bladder urothelial carcinoma, squamous cell carcinoma,Adenocarcinoma and adjacent normal tissue5cases each) were measured by Western blotting, and analysed the expression of HER2protein in bladder canner of different histological type.Results1.The expression of HER2in bladder cancer tissues has no significant correlation with sex and age(P>0.05). The positive expression of bladder tumor tissues (72.60%,73/53) was significantly higher than that in normal bladder tissue (7.69%,13/1), The difference was statistically significant (P<0.05). The positive expression of Squamous cell carcinoma and adenocarcinoma of bladder was higher than that of bladder transitional cell group, The difference was statistically significant (P<0.05). The positive expression rate of HER2in stage0-I is46.66%,that in stage â…¡ is72.72%, that in stage III is88.00%,, the positive rate of HER2expression was significantly higher With stage increased. The positive expression rate of HER2in High differentiated tissue is55.56%,that in Medium differentiated tissue is85.71%, that in Low differentiated tissue is93.75%, the positive rate of HER2expression was significantly higher With reducing the degree of differentiation.2the expression of HER2were significantly higher in bladder cancer than that of controls. Compared with the bladder urothelial carcinoma, HER2protein in squamous cell carcinoma and adenocarcinoma of bladder tissue of bladder significantly increased, The difference was statistically significant (P<0.05),Compared with adenocarcinoma of bladder tissue, HER2expression in the bladder squamous cell carcinoma is higher, but the difference was not statistically significant.ConclusionsThe expression of HER2in bladder squamous cell carcinoma or adenocarcinoma tissues was significantly higher than that in bladder urothelial carcinoma; The expression of HER2in bladder cancer tissues has no significant correlation with sex and age, closely correlated with the TNM staging and differentiation.
