| Background and objectiveSince ancient times, trauma is harmful to human health, which is increasing along with the social development and human progress. Frequent occurrence of daily life, traffic accidents, natural disasters, and serious situations can cause disability, even life threatening security. At present, in the process of wound healing, a lot of methods promote wound healing, including traditional Chinese medicine and Western medicine, traditional dressing and new dressing, skin and cell membrane transplantation, gene therapy, stem cell therapy, cytokine. It is said that in wound healing of traditional Chinese medicine, essence of soft tissue injury is blood stasis, context damage after injury, blood from the meridians and into stasis, stasis and heat, so the treatment is mainly to promoting blood circulation to remove blood stasis, heat and cooling blood. Compound traditional Chinese medicine ointment used in this experiment is from rhubarb, coptis root, garden burnet, beeswax, borneol, halloysite, calamine, Cortex Moutan, Galla chinensis, three seven, frankincense, myrrh, sesame etc. At present, the experiments and reserchs of compound traditional Chinese medicine ointment on porcine split-thickness skin wound healing are less. Therefore, the study on the effects of drugs in the process of wound healing, by detecting healing time, healing rate, the thickness of epidermis, body temperature, and the content of transforming growth factor beta, catalase, malondialdehyde and WBC. Materials and methodsThe experiment used114-month old health white pigs, weight30~35kg, irrespective of genders, which were provided by the experimental animal center of Henan Technology University, and varieties of the United States Duroc. Prepared the porcine split-thickness skin wound model, and the depth of the wound is to the deep dermis, a depth of about3/4of the dermal thickness. The experiments were separated into compound traditional Chinese medicine ointment group (group A), Silikai nanometer silver antibacterial gel group (group B), blank control group (group C), treated respectively with compound Chinese medicine ointment, Silikai nanometer silver antibacterial gel, normal saline gauze, and quantitative greased. Everyday observe the wound healing time and the healing degree on different time of the three groups. Monitor porcine temperature. Every5days after injury obtained the wound tissue of three groups, and tested the porcine skin transforming growth factor beta (TGF-β) content, epidermal thickness, catalase (SOD), malondialdehyde (MDA), total protein concentration. The samples were stained by HE, observed the change of epidermal and dermal. Tested white blood cell values took from the ear vein every5days.Results1In the animal experiments, the healing time of A group, B group and C group, were respectively13.17±.75d,18.42±1.93d,24.67±2.64d, P<0.05.2In comparison of wound healing rate, A group was higher than B group and C group on seven day, fourteen day, and twenty-one day after injury, P<0.05.3In comparison of TGF-β content on5-day after injury, group A was196.36±1.53pg/ml,160.52±1.43pg/ml in B group, and150.65±3.60pg/ml in C group. A group was higher than B group and C group, P<0.05. On9-day after injury, A group was183.39±1.73pg/ml,157.19±3.58pg/ml in B group,166.68±6.50pg/ml in C group. A group was higher than B group and C group, P<0.05. On other time points A group was lower than B group and C group.4In comparison of the wound epidermis thickness on5-day to24-day after injury, the A group was higher than that in B group and C group, P<0.05. But on29-day after injury, P>0.05.5In comparison of the total protein content of each group in wounds on5-day to24-day after injury, the A group was higher than that in B group and C group, P<0.05. But on1day before injury and29-day after injury, P>0.05.6In comparison of SOD content in each group on5-days,9-day,14-day after injury, A group was higher than B group and C group, P<0.05. But on lday before injury and29-day after injury, P>0.05.7In comparison of MDA content in each group on5-day to24-day after injury, the A group was lower than that of B group and C group, P<0.05. But on1day before injury and29-day after injury, P>0.05.8Within30days of wound dressing, the temperature of group A was not higher than39℃. The temperature of group B on the seventh day was higher than39℃, and other time was not higher than39℃. The temperature of group C was higher than39℃on3-day to6-day, and other time was not higher than39℃.9In the process of wound healing, the curves of leukocyte value showed that the value of the A group and B group floated in the normal range, and C Group floated high, especially on5-day to24-day after injury, higher than the normal value, and up to41.8X109/L on5-day after injury.Conclusions1Preparation of porcine split-thickness skin wound model is reliable and stable.2Compound traditional Chinese medicine ointment in porcine split-thickness skin wound healing process, can shorten the wound healing time, raise the rate of wound healing, and increased the content of TGF-β in wound tissues, promote wound healing.3Compound traditional Chinese medicine ointment can improve the SOD content in the wound tissue, enhance antioxidant capacity of drug; reduce the content of MDA in the wound tissue, reduce the cell toxicity. Local or systemic inflammatory response caused by reducing skin damage, anti-inflammatory action.4Compound traditional Chinese medicine ointment reduce exudate in wound tissues, embody the exudation function. |
