Objective:To compare with the influence which rendering in the corresponding hygroscopicity of dry, dry extract yield and different chemical composition in dry extract when chitosan flocculation clarifying technique and alcohol sink using in refined for the water solution of Herba lysimachiae, Hedyotis diffusa Willd and Fructus toosendan.Methods:Contrast two methods from yield, and output of sugars, amino acids and content of effective compositions, and the residual of protein and tannin, and hygroscopicity of the. Using UV-spectrophotometric method to determine the content sugars, amino acids, protein and tannin in the dry extract from the three kinds of traditional Chinese medicine processed by the two different methods. Using HPLC to determine the content of quercetin, ferulic acid and grass fingerprint which in the dry extract from Herba lysimachiae and Hedyotis diffusa Willd. Using the constant-temperature-humidity way and constant-temperature-change-wet way to determine the moisture absorption curves and the critical relative humidity in the dry extract from the three kinds of traditional Chinese medicine processed by the two different methods.Results:The three herbs’access rate of the dry extract is significantly higher from chitosan flocculation clarifying method to the alcohol method, it’s similar in retention of quercetin and ferulic acid from Chitosan flocculation clarifying method to alcohol sink method. Chitosan flocculation clarifying method is stronger than alcohol sinking method in removaling efficiency sugars, amino acids and tannin, while similar in removaling proteins. The hygroscopic of dry extract made by chitosan flocculation clarifying method is low relatively then by hygroscopic alcohol sinking method. Through the analysis and comparison of the Herba lysimachiae and Hedyotis diffusa Willd’s fingerprint, chitosan flocculation clarifying method can keep Herba lysimachiae hydrophilic components better while alcohol sinking method can keep Hedyotis diffusa Willd hydrophilic compounds better. So we can know that there is no fixed relationship in hydrophilic component retention of the two processing methods, it has randomness.Conclusion:The experimental data could be used to provide theoretical support and experimental basis of a further research for the mechanism which chitosan flocculation clarifying method can reduce dry extract medicine moisture absorption. As well as giving an experiment basis to the using of chitosan flocculation clarifying method in the preparation process.
