Cyrtomiwn fortunei J.Sm. is a kind of perennial herbaceous plant which belongs to Cyrtomium of Dryopteridaceae. It is widely distributed over China. And there are abundant resources. Cyrtomium fortunei is one of the original plant of GuanZhong which is a kind of traditional Chinese medicine. In this paper, the ultrasonic extraction process of total flavinoids from the underground part of Cyrtomium fortunei was optimized. The antioxidative activites of different polarity were measured. The hypoglycemic effect of ethyl acetate fraction on the diabetic mice induced by alloxan was studied. This provides a basis for the development and utilization of Cyrtomium fortunei resource. The main experimental results are as follows:(1)The content of total flavonoids was measured by NaNO2-Al(NO3)3colorimetric with rutin as standard control. The influences of the factors such as ethanol concentration,extraction time and the ratio of row materials to solution on the extraction rate of the total flavonoids in the underground part of Cyrtomium fortunei were discussed. And the optimal process was confirmed by single and orthogonal experiments and listed as follows:the ethanol concentration of60%, the extracting time of30min and the ratio of row materials to solution of1:40(g/mL). In this condition, the extraction rate of total flavonoids was1.34%. The influence of ethanol concentration and extracting time on the extraction rate of the total flavonoids were significant difference at the0.01level. And the influence of the ratio of row materials to solution on the extraction rate of the total flavonoids was significant difference at the0.05level. The influence order of the three factors was th@extracting time>the ethanol concentration>the ratio of row materials to solution.(2)The content of flavonoid in the different polarity parts from the underground part of Cyrtomium fortunei was determined. The results show that the content of flavonoid in ethyl acetate fraction is52.97%which is the highest among the4different polarity parts. The content of flavonoid in water saturation normalbutyl alcohol fraction is16.06%which is higher than chloroform fraction and water fraction. And the content of flavonoid in chloroform fraction and water fraction is6.19%and4.28%. According to the scavenging activity for DPPH radicals and reducing activity, the antioxidant activities of different polarity fractions were investigated. The results show that four fractions all had different antioxidant effects and existed concentration-effect relationships in some extent. The scavenging activity for DPPH radicals is ethyl acetate fraction> water saturation normalbutyl alcohol fraction> water fraction> chloroform fraction>BHT. When the concentration of different fractions are less than or equal to0.4mg/mL, the ability to reduce ferric ions is as follows:ethyl acetate fraction>water saturation normalbutyl alcohol fraction> water fraction>BHT> chloroform fraction.(3)The diabetic mice model were induced by injecting intraperitoneally with alloxan. To study the effect of the ethyl acetate fraction in which the content of flavonoid is52.97%on the blood glucose and plasma lipid of diabetic mice by testing the concentration of TG, TC, LDL-C and HDL-C. The ethyl acetate fraction has effect on improving the blood lipid of diabetic mice induced by alloxan. The result proved that the ethyl acetate has alleviated effect on diabetic.
