| Atherosclerosis (AS), as the pathological basis of cardiovascular diseases,is a common disease that harms lives. Many hypotheses have been proposed bydomestic or overseas scholars in order to expound the mechanism of it. Amongall these propositions,“damage-response theory and the subsequently-modifiedinflammation theory†are commonly recognized. And among all the factors causingdamage-response, the anomaly of lipid metabolism is of great importance, hencethe rectification of such abnormalities is regarded as the key approach in theprevention and treatment of AS. In recent years, although they are adopted inorder to achieve such an effect, the western medicines are in generalfunctionally single and highly dependent with too many side-effects. Incomparison, the traditional Chinese medicine which exerts its efficacy throughpattern identification is of such characteristics as treatment according to eachindividual, holism and slight side-effects. My tutor believes that the mainpathomechanism of AS is the biding of phlegm and stasis, the two of which arepathological products caused by fluid abnormality and can turn into each other,therefore in treatment, the focus should be given to both of them, namelysimultaneous treatment of phlegm and stasis should be adopted. This experimentapply such principle to formulate a recipe, Channel-unblocking for RemovingTurbidity Decoction (通脉化浊汤,TÅng Mà i Huà zhuó TÄng, abbrevated as CRTD),and by observing its mechanism on affecting the AS rats, the role played byprinciple of “simultaneous treatment of phlegm and stasis†in treating AS isdiscussed, and experimental foundation are concluded for clinical treatment ofAS through TCM therapy.Purpose:AS rats are induced through calcium overload and lipid infiltration.Myocardial ischemia (MI) model are induced by intraperitoneal injection ofpituitrin. Observe CRTD in each dosage and Wenstardin’s effect on AS rats. Method:1. Group and model induction: Rats suitable for the experiment are randomizedinto blank group, model group, CRTD high dose group, CRTD middle dose group,CRTD low dose group and control group of Wenstardin. The rats form model groupand each treatment group are feed with high-lipid chow from3days after thelavage of Vitamin D3(700thousand UI/KG) on. Rats from the blank group are lavegedby normal saline of the same volume for3days and then feed with normal chow.12weeks later,3rats are randomly sampled in each group except the blank oneto observe the induction results.2. Intervention: After induced, the models from each group are given interventiontreatment with medicinals with volume according to their weight (5ml/kg). Therats from blank group are given normal saline of same volume with their body.They are sacrificed to take sample in the7th,14th and28th day.3. Materials and examinations: Before the each sampling, the rats from modelgroup and treatment group are given intraperitoneal injection of pituitrin tocause them get MI according to their weight (30U/kg), while the rats from blankgroup are given normal saline of the same volume. On the day of sampling, randomlyselected rats are injected with25%urethane into their abdominal cavities andblood is sampled from their aorta abdominalis to be prepared as serum lipid (TCand TG), lipid protein (HDL and LDL), apolipoprotein (Apo), endothelin (ET),thromboxane A2(TXA2) and prostacyclin(PGI2). Then cut open the abdomen and stripoff the aorta to prepare for being stained by oil red O and Microscopicobservation.Results:1. AS and MI models can be successfully induced through the methods of lipidinfiltration, calcium overload as well as intraperitoneal injection ofpituitrin;2. CRTD serves to decrease notably the levels of TC, TG, LDL-C and apoB100, andincrease those of HDL-C and apoAI;3. Reduced levels of TXB2and ET and raised level of6-k-PGFla can be observed in CRTD treatment group;4. The simultaneous treatment of phlegm and stasis is currently the main methodand plays a key role in treating CHD.Conclusion: CRTD, the recipe that is formulated under the method ofsimultaneous treatment of phlegm and stasis, can remove lipid plaque, Repairdamaged endothelium, prevent thrombosis and relieve the MI through itsregulation on lipid metabolism, so as to achieve the efficacy on AS. |
