[Objective]Peripheral nerve injury is a common clinical disease, and sciatic nerve injury is a typical Peripheral nerve injury. In this experiment, make the sciatic nerve clamping injury model to simulate clinical sciatic nerve injury, and timing given by Electro-acupuncture qualitative and quantitative treatment. From the sensory-motor transduction pathways explore the mechanism of Electro-acupuncture treatment in sciatic nerve injury, and through the expression of GAP-43study the effective factor, the onset of ways and time by Electro-acupuncture, to Provide strong scientific evidence for Electro-acupuncture treatment in clinic and improve the Patient’s symptoms and prognosis.[Methods]Using SD rat sciatic nerve clamping injury model, the study used Electro-acupuncture simulator on GB30,UB37, BL57and GB34. And also, solar-thermal pain threshold resistance and inclined plate test is used from a behavioral point of view to explore the recovery on sensory and motor function after therapy. Through observing the variation of the ventral horn of the spinal cord, dorsal root ganglia, damage points nervous, neuromuscular junction and muscle, the promotion of therapy’s effect on neuronal regeneration is sought on aspect of morphology. Through the research on changes of GAP-43expression in the key parts, approach and mechanism are revealed by electro-acupuncture on sciatic nerve injury.[Results]1. EthologyThe inclined plate test results show that7days after modeling, model group were significantly different (P<0.05) compared with the normal group, after sciatic nerve injury in rats motor function and muscle strength is greatly affected successful model.10times after Electro-acupuncture treatment, the results of electro-acupuncture treatment group is better than model group and model control group (P<0.05), and close to the level of the normal group.20times after Electro-acupuncture, Electro-acupuncture treatment group close or exceed the level of the normal group, and there is still a significant difference (P<0.05) with the model group and model control group. It means that Electro-acupuncture can be effective in promoting the recovery of motor function and lower extremity muscle strength of the sciatic nerve injury in rats.Solar-thermal pain threshold resistance results are as follows:after modeling7days, Electro-acupuncture treatment10times and20times, the results of left (contralateral) among the groups is no difference;7days after modeling the results of the right side (ipsilateral), compared to the model group and the normal group were significantly different (P<0.05). It means that after sciatic nerve injury in rats sensory function is greatly affected.10and20 times after Electro-acupuncture treatment, the results of Electro-acupuncture treatment group were lower than the model group and model control group (P<0.05), the hyperalgesia scores of Electro-acupuncture treatment group was significantly decreased compared with model group (P<0.05), It means that Electro-acupuncture can Promote the recovery of sensory function of the sciatic nerve injury in rats.2. MorphologyThe results of gastrocnemius wet weight are as follows:7days after modeling, electro-acupuncture treatment10and20times, there were no differences between the results of left gastrocnemius muscle wet weight.7days after modeling, the results of sham operation group and model group were reduced to some extent, and the model group is more obvious. The results of the gastrocnemius muscle wet weight and gastrocnemius muscle recovery rate have significant difference between the model group, normal group and sham group (P<0.05).10times after electro-acupuncture, the results of electro-acupuncture treatment group increased, and there was a significant difference between the model group and model control group(P<0.05), but the wet weight and recovery rate of rats in each group as compared with the normal group there are still differences (P<0.05).20times after electro-acupuncture, the results of electro-acupuncture treatment group are still higher than the model group and model control group, but still a gap compared with the normal group (P<0.05).7days after modeling, right gastrocnemius muscle cell diameter of model rats was significantly less than the normal group and sham operation group, and the difference was statistically significant (P<0.05).10times after electro-acupuncture, the results of electro-acupuncture treatment group increased to some extent, and there is a significant difference compared with the model group and model control group (P<0.05), but compared with the normal group there is a gap (P<0.05).20times after electro-acupuncture treatment, the results of each group are gradually increased, and the electro-acupuncture treatment group was significantly better than the model group and model control group (P<0.05), but has not yet reached the level of the normal group.HE results of dorsal root ganglion:Under normal circumstances, the majority of neurons in the dorsal root ganglion was round, larger, cytoplasmic eosinophilic, large round nucleus, basophilic staining shallow, clear nuclear and distribution of satellite cells around the cell body; model group neuronal cell swelling, deformation and dissolution, neurons in different degrees of necrosis, cell morphology sizes, uneven cytoplasmic staining; through electro-acupuncture, neural the element cell swelling and deformation is not obvious that the cell body shape more regular, mostly circular.HE in the spinal cord:Under normal circumstances, the ventral horn of the spinal cord neurons arranged in neat rows, the nucleus is located in the center of the cell, the nucleolus is clear, evenly distributed Nissl bodies; spinal cord ventral horn neurons of model group irregular arrangement, reducing the number of cell swelling and degeneration Phenomenon, lighter staining nuclei towards the edge and the Nissl bodies dissolved disappear; through electro-acupuncture, spinal cord ventral horn neuronal degeneration, damage to a lesser extent, arranged more regular Part of the nucleus toward the edge and small amount of nuclear regeneration.The HE results of gastrocnemius:Under normal circumstances, the gastrocnemius muscle in the same size, arranged in neat rows, clear nucleus, cytoplasm uniform and moderate muscle fiber gap; diameter of the gastrocnemius muscle cells of the model group was significantly reduced, the uneven distribution of the nucleus is clear, but the number is significantly increased, muscle. fiber gap increased and shrank Phenomenon; through electro-acupuncture treatment, diameter of gastrocnemius muscle cells uniform, clear nucleus, to a lesser extent of muscle fiber atrophy.HE results of sciatic nerve:Under normal circumstances, the axons of neurons and schwann cells arranged in neat rows; In model group, Part of the axon and schwann cells arranged in irregular swelling, ulceration, or even crack, and macrophages, glial cells appear obvious Proliferation; after electro-acupuncture treatment Schwann cells and axons are basically normal.Silver stain results of motor end Plate:Under normal circumstances, the motor end Plate was brown or black, clear, smooth edges, oval, muscle texture clear; uniform coloring between the various Parts, structural contrast, no shrinkage Phenomenon; in model group, motor end Plate uneven distribution of smaller size, connective tissue Proliferation significantly, fuzzy, Pale staining,,and Part of the motor end Plate disintegration, only remnants can be seen, the remnants of a large number of nuclei accumulation; through electro-acupuncture, motor end Plate gradually formed, remnants gradually reduced.3.
