ObjecriveThis topic through HE staining observation myelinated axon counts and WB detect nuclear factor p50、p65,which observe the effect of electro acupuncture combined with nerve loosening on the axonal regeneration and p50、p65 of the serum after sciatic nerve compression injury in rabbits.To investigate the molecular mechanism of the regulation of axonal regeneration after sciatic nerve injury in rabbits by electro acupuncture combined with nerve loosening,Thereby, it can provide the theoretical basis for the treatment of sciatic nerve injury by electro it can provide the theoretical basis for the treatment of sciatic nerve injury byelectro acupuncture combined with nerve loosening.Methods1.18 New Zealand rabbits were prepared using clamp Ⅲ Sunderland degree injury of rabbit sciatic nerve compression model, Were randomly divided into 4 groups:model group, electro acupuncture combined with nerve mobilization (group Zhen Song); Another 6 rats were established to establish the sham operation group, which was exposed to the sciatic nerve without nerve compression.3 days after surgery, Zhen Song group:after the first clip of spinal acupuncture (EA) after nerve loosening (all treatment by one person); BDNF group:intramuscular injection of BDNF. The groups were 1 times/d, Weekly treatment for 6 days, a total of 4 weeks. The model group and the sham operation group were not intervened. After 4 weeks treatment, the number of the spinal cord axons and the Blot Western were detected by HE staining, and the nuclear factor P50 and p65 were detected by staining.Result1.HE staining was used to observe the growth of axons of the sciatic nerve:After 4 weeks of treatment, After 4 weeks of treatment, Zhen Song group, BDNF group and sham group neurite outgrowth, nerve fibers, bundles membrane structures are better than the untreated group; The axonal growth, nerve fiber and bundle membrane structure of the Zhen Song group were better than BDNF groups.2.P50、P65, After 4 weeks of treatment, After 4 weeks of treatment, P50, P65 expression in each group were higher than sham group,the difference was statistically significant (P<0.05).Zhen Song group, BDNF group P50, P65 compared with the model group, the difference was statistically significant (P <0.05).Zhen Song groupP50, P65 slightly higher than BDNF expression group, but not statistically significant (P> 0.05).Conclusion1. Jiaji Electro acupuncture combined with nerve mobilization and BDNF can promote axonal regeneration after sciatic nerve injury.2.Jiaji Electro acupuncture combined with nerve mobilization and serum BDNF can make serum nuclear factor P50, P65 content increased.3.Jiaji Electro acupuncture combined with nerve mobilization promote axonal regeneration after peripheral nerve injury may be associated with the expression of P50, P65 increase.
