[Objective] Study of different concentrations of Shenqi-fuzheng injection on in vitro cultured human breast cancer MDA-MB-231cells inhibit proliferation and induce apoptosis.To study the effects of different concentrations of Shenqi-fuzheng injection on proliferation and apoptosis of basal-like breast cancer line MDA-MB-231cells in vitro.[Methods] There were seven groups divided in this study:One control group (MDA-MB-231cells cultured without Shenqi-fuzheng injection) and six experimental groups in which MDA-MB-231cells were cultured at different dose (A:1.25g/L, B:2.5g/L, C:5g/L, D:10g/L, E:20g/L, F:40g/L). Proliferation of MDA-MB-231cells were evaluated with MIT assay, including cell morphology, cell growth curve and cell inhibition rate. Cell cycle distribution and apoptosis of MDA-MB-231cells induced by Shenqi-fuzheng injection were mesured by flow cytometry instrument technology (FCM).The cultured human breast cancer MDA-MB-231cells were divided into blank control group and6different concentrations of Shenqi-fuzheng injection groups, carry through inhibition experiment in vitro. The human breast cancer MDA-MB-231cells as the research object, Shenqi-fuzheng Injection for the treatment of factors.With MIT-assay method, determinate of different concentrations of Shenqi-fuzheng injection on MDA-MB-231cells proliferation effect after different time,including cell morphology, cell growth curve, cell inhibition rate, analysis of dose-effect and time-effect relationship. And using flow cytometry instrument technology (FCM) determination of MDA-MB-231cells, the change of cell cycle distribution and apoptosis.[Result] Shenqi-fuzheng injection can inhibit the growth of the MDA-MB-231cells, compared with the blank control group was statistically difference (P<0.01) and the most significant inhibitory effect was at48h cultured, And the inhibition effect was dose dependent. Flow cytometry results showed that Shenqi-fuzheng injection can inhibit the MDA-MB-231cells at S phase (compared with the blank control group was statistically difference, P<0.01), and induce apoptosis, which was dose dependent.[Conclusion] Shenqi-fuzheng injection can significantly inhibit the proliferation of the human breast cancer MDA-MB-231cells in vitro, and induce cell cycle arrest and apoptosis, which showed dose and time dependence The results indicate that Shenqi-fuzheng injection may bring therapeutic benefits for basal-like breast cancer patients.
