Objects:We built up an exercise protocol of one week endurance exercise, using the DHT slow-release pellets, to explore the influence of androgen and exercise on Akt/GSK3p expression in rat’s skeletal muscle.Methods:24male SD rats aged8weeks were randomly divided into four groups, six for each:group control (group C), group exercise(group E), group silence plus DHT(group CD), group exercise plus DHT(group ED). Group CD and group ED underwent a surgery with neck-embedded DHT slow-release pellets, while group C and group E were taken a sham. Group E and group ED had a treadmill training for one week (5%,20m/min,60min/d), while group C and group CD were fed in scilence.12hours after the last training, white muscle of the right gastrocnemius were taken. Phosphorylation expression of Akt(Ser473), GSK3β(Ser9) and eIF2Bε(Ser535) and total protein were tested. Western blot were adopted to test the phosphorylation expression and Bradford method for total protein.Results:(1)Compared with group C, total protein of gastrocnemius muscle in group ED significantly increased(P<0.05), and in group E and group CD, total protein trended to increase.(2) Compared with group C, there existed a trend to increase phos-Akt(Ser473) and phos-GSK3β(Ser9) in group E, and phos-eIF2Bε(Ser535) trended to be depressed. Group ED showed the similar changes when comparing with group CD.(3) Phos-Akt (Ser473) and phos-GSK3β (Ser9) trend to increase and phos-EIF2Bε (Ser535) was significantly decreased (P<0.05) in group CD(compared with group C) and group ED (compared with group E).(4) Compared with group C, phos-Akt (Ser473) and phos-GSK3β(Ser9) in group ED were significantly increased (P <0.01), and phos-eIF2Bs (Ser535)was significantly reduced (P<0.01).Conclusion:(1) Androgen and exercise can cause a trend to promote phos-Akt (Ser473) and phos-GSK3P(Ser9) in skeletal muscle,and androgen had a more significant inhibitory effect on phos-eIF2Bε (Ser535).(2) Androgen enhances the increasing effect of exercise on skeletal muscle protein, which may involve Akt(Ser473)/GSK3β(Ser9)/eIF2Bε(Ser535) pathway.
