Objecties:This study attempts to ApoE-/-mice as the research object, the NO signal transduction mechanisms as clues, the aerobic exercise as the main interventions.For researching Exercise intervention and dynamic atherosclerosis formation, try to explore between aerobic exercise and NO signal transduction mechanism.Methods:By10weeks of age C57BL/6J of ApoE (-/-) mice,48, Male and Level of SPF, body weight (18.24+0.60) g. Were randomly divided into Quiet group (control group), exercise group, medication group (pravastatin sodium tablets),16rats in each group. After12weeks of intervention,Three groups of (ApoE-/-) mice are detected by serum blood detection, Reverse transcrition polymerse-chainreaction(RT-PCR), enzyme-linked immunosorbent assay (ELISA)and HE staining of thoracic aortic sections to observe the morphological changes. Which Cytokines Cholesterol (TC), triglyceride (TG), high density lipoprotein (HDL), low density lipoprotein (LDL)(nitric oxide) NO, endothelin (ET), calcitonin genepeptide (CGRP) of NOS (nitric oxide synthase) Were detected in serum, the other in thoracic aorta--(C-reactive protein) CRP,(Janus kinase)JAK,(signal-transduction-and-transcription-activator)STATmRNA.These related cytokines were detected with the NO signal transduction by above experimental techniques.Results:①erobic exercise group, treatment group (pravastatin sodium tablets) in the lipid composition of the TC, TG and LDL content were statistically significant compared to the control group; where the TC, TG and LDL detection results showed that the exercise group and treatment group was no significant difference; the aerobic exercise group serum HDL levels were lower than the control group, treatment group (P<0.05); NO detection results showed that serum NO levels in the exercise group was significantly higher than the control group (P<0.01),and higher than the treatment group (P<0.05).②By HE staining of ApoE-/-mice thoracic aorta morphological observation, the results showed that aerobic exercise can significantly reduce the plaque of the aorta.③ELISA to determine serum concentration of endothelin (ET) levels was to the highest in the control group, treatment group, sports group decreases; control group compared to treatment group and sports group, the difference was statistically significant (p<0.01), the exercise group and treatment group was no significant difference (p>0.05). The quiet group were the lowest in these groups for the concentration of serum calcitonin (CGRP); compared with other two groups,the difference was statistically significant (p<0.01), while the exercise group and treatment group differences were statistically significance (p<0.05). The exercise group were the highest in these groups for Nitric oxide synthase (NOS) in serum levels,treatment group and control group decreases; and exercise group compared with treatment group and control group, the differences were statistically significant (p<0.01).(4) By RT-PCR assay the JAK mRNA expression levels to the highest in the control group, the lowest treatment group, and exercise group compared with treatment group and control group, the differences were statistically significant (p<0.05). CRP mRNA expression levels to the highest in the control group,and exercise group and treatment group with brightness was no significant difference, but are weaker than the control group. STATmRNA expression levels to the highest in the control group, treatment group is weaker, followed by the exercise group is the weakest; the exercise group and control group difference was statistically significant (p<0.01), the treatment group compared with control group difference was statistically significant (p<0.05).Conclusion:①By HE staining of ApoE-/-mice thoracic aorta morphology observed, indicating that the aerobic exercise intervention significantly reduced aortic plaque formation.②Through the comparative analysis of lipids on the subjects and the experimental results of NO-related cytokines, that aerobic exercise and administration (pravastatin sodium tablets) can to some extent on the ApoE-/-mice atherosclerosis formation, both are the role of benign intervention, but the experimental results showed that both cytokines degree of intervention different.③groups of ApoE-/-mice sera showed that aerobic exercise and medication can lower blood lipid composition, the experiments showed that aerobic exercise can increase serum NO, NOS and CGPR in the content, reducing the content of the ET; For mRNA expression of the thoracic aorta, aerobic exercise can inhibit the expression of CRP, STAT and JAK, and thus to as the formation of the interventionist role to alleviate atherosclerosis (AS).
