Objective: In order to approach the influence of aerobic training on the alternation of lipid metabolism and the other possible mechanisms beyond lipid-regulation of aerobic training in anti-atherosclerosis,we observed the influence of aerobic training on the formation of atherosclerotic lesions, the concentration of TC, LDL,HDL,HDL2,HDL3 and nitric oxide(NO) in serum, the activity of superoxide dismute(SOD) in serum and liver, the content of inducible nitric oxidase(iNOS) and endothelial nitric oxidase(eNOS), the lipid peroxidation in thoracic aorta in ApoE-deficient mice.Methods:the atherosclerotic lesions,the serumal TC,LDL,HDL, HDL2.HDL3 and NO,the SOD activity in serum and liver, the content of thoracic aortic iNOS and eNOS by ABC immunohistochemical staining and lipid peroxidation by Cold Schiff histochemical method combined with computer image quantitative analysis technique on frozen sections of thoracic aorta were measured. Harris'hematoxylin(HE) staining on frozen sections of thoracic aorta.Oil Red O(ORO) staining on the intima of the thoracic aorta.ResultsrCompared with controls, aerobic training delayed the plaque formation in ApoE-deficient mice (P<0.01); the concentrations of serumal TC,LDLwere significantly increased(P<0.01),while the alternations of HDL, HDL2 and HDL3 were insignificant (P>0.05) ; the concentration of serumal NO was increased(P<0.0l); the activity of SOD in serum was increased (P<0.05) ,while that of SOD in liver was increased insignificantly(P=0.056); the content of thoracic aortic iNOS was decreased (P<0.05) and that of eNOS was increased (P<0.01); the lipid peroxidative reaction of thoracic aorta was weaker (P<0.01);the lesions area in thoracic aortic wall and intima was found less by gross light microscopic examination.Conclusions:The lipid levels were significantly increased,while the anti-oxidation function and endothelial function were significantly increased, atherosclerotic plaques were significantly decreased after 10-week aerobic training in ApoE-deficient mice, This may reflect a new mechanism of aerobic training on the anti-atherosclerosis,independent of lipid-regulation mechanism.
