Object:This experiment on skeletal muscle apoptosis by medium、 high-intensity aerobic exercise, the expression of the movement on TRPC1mRNA and TRPC1protein concentration, discussed the role of TRPC1in muscle cell apoptosis, the process of sports training, sports rehabilitation, sports therapy, gene therapy to provide a theoretical basis.Methods:30healthy male SD rats aged six weeks, according to body weight were divided into normal control group (NC, n=10), moderate-intensity aerobic exercise group (ME, n=10), high-intensity aerobic exercise group (HE, n=10). ME group, gradient5°,19.3m·min-1×60min; HE group gradient10°,26.8m·min-1×40min. NC group and aerobic exercise group rats were at rest or after exercise at12hours after ether anesthesia were decapitated from gastrocnemius and soleus, respectively. Using HE staining to identify changes of skeletal muscle in cell morphology; TUNEL was detected apoptosis in skeletal muscle; the calcium content of skeletal muscle cells of the colorimetric method; by RT-PCR detect TRPC1mRNA expression; Western blotting to determination the content of TRPC1protein in skeletal muscle cells.Results:(1) After8weeks of moderate-intensity, high-intensity aerobic exercise training, the soleus and gastrocnemius have suffered varying degrees of muscle fibers arranged in irregular, such as the gap widened between the muscle fibers, high-intensity aerobic exercise group flounder muscle is particularly obvious.(2) Compared with normal control group, moderate-intensity aerobic exercise group, the apoptosis rate of the high-intensity aerobic exercise group significantly increased, the apoptotic index was significantly increased (P<0.05).(3) Compared with normal control group, moderate-intensity aerobic exercise group of high-intensity group of rat gastrocnemius and soleus muscle of Ca2+content was significantly increased (P<0.05); compared with the moderate-intensity aerobic exercise group, large intensity exercise group gastrocnemius, soleus muscle Ca2+of the content were significantly increased (P<0.01).(4) Compared with normal control group, the moderate-intensity aerobic exercise group of high-intensity group rat gastrocnemius and soleus TRPC1protein content increased.Conclusions:(1) High-intensity aerobic exercise on skeletal muscle cell apoptosis, in particular a greater impact on the soleus muscle cell apoptosis.(2) Compared with N、ME group, the apoptotic index of the gastrocnemius increased in HE group, statistically significant (p<0.05); Compared with NC group, the apoptotic index of the soleus muscle increased in ME group, statistically significant (p<0.05); Compared with NC group, the apoptotic index of the soleus muscle increased in HE group, statistically significant (p<0.01); Compared with ME group, the apoptotic index of the soleus muscle increased in HE group, statistically significant (p<0.05).(3) Compared with NC group, Ca2+content of gastrocnemius increased by37.8%in ME group, statistically significant (p<0.05), Ca2+content of gastrocnemius increased by87.9%in HE group, statistically significant (p<0.01); Compared with ME group, Ca2+content of gastrocnemius increased by36.4%in HE group, statistically significant (p<0.05). Compared with NC group, Ca2+content of soleus increased in ME group, statistically significant, statistically significant (p<0.05), Ca2+content of soleus increased by113.6%in HE group, statistically significant (p<0.01);Compared with ME group, Ca2+content of soleus increased in HE group, statistically significant, statistically significant (p<0.01).(4) Compared with NC group, TRPC1mRNA、 TRPC1、TRPC1/B-actin ratio of the gastrocnemius increased in HE group, statistically significant (p<0.01); Compared with ME group, TRPC1mRNA、TRPC1、TRPC1/B-actin ratio of the gastrocnemius increased in HE group, statistically significant (p<0.05).
