Objective: to exercise related diseases caused by high intensity exercise is one of the most important diseases in outpatient department of sports hospital, this study explored the effects of different intensity exercise on rat skeletal muscle morphology and apoptosis,from morphology Study of observation, Content of superoxide dismutase (SOD) and malondialdehyde (MDA) in skeletal muscle tissue, have an important role in the skeletal muscle injury caused by high intensity exercise on the prevention and treatment of sports injury and at the same time, movement disorders, and provide a theoretical support for the cause of high intensity exercise.Methods: 30 healthy male Sprague Dauley (SD) rats aged in March were randomly divided into three groups: the control group (n=10), the general aerobic exercise group(n=10) and the long time intensive exercise group (n=10). The quiet control group without swimming exercise, general aerobic exercise group and high intensity exercise for a long time will reference experimental method by Hu Yazhe et al, Tian Zhenjun,animal model by swimming exercise system. General aerobic exercise group and a long time high-intensity exercise group training cycle were 10 weeks, adaptive swimming for 2 weeks, 6 times / week. Rats swimming time: the first 10min, after each increase of 5min, until the time rose to 60min. From the beginning of the third week, the general aerobic exercise group daily swimming training (60min body weight); long time high intensity exercise group weight training, third, 4, 5 weeks, weight were about 1% of body weight, 2%, 3%, the swimming time was 120min. The weight at 6-10 weeks was about 6% of the body weight of rats, and the swimming time was 150min. After 10 weeks, the rats were anesthetized with gastrocnemius muscle, and then stained with HE and TUNEL.Meanwhile, skeletal muscle homogenate was prepared to detect the content of SOD and MDA in skeletal muscle tissue.Results: (1) Skeletal muscle HE staining results: Control group: Normal skeletal muscle structure, muscle fibers arranged closely, regular shape, muscle membrane clear and complete, clear and regular muscle strips. Generally aerobic exercise group: muscle fiber thickening. Lengthy time high intensity exercise group: severe injury at the fragment of disordered structure fuzzy, is the result of cell lysis.(2) The results of TUNEL: The positive expression of quiet control group showed a small amount of brown nuclei, the general aerobic exercise group had more positive expression of the nucleus, the positive expression showed more long time high intensity exercise group and control group of the nucleus; Compared with the control group, the value of the integrated optical density (IOD) was increased, and the value of IOD in the aerobic exercise group and the long time high intensity exercise group increased significantly (P < 0.05);and the general aerobic exercise group IOD values than the long time, high intensity exercise group IOD value increased significantly (P < 0.05).(3) The results of MDA content of SOD, MDA: General aerobic exercise group compared with control group, P < 0.05, there was significant difference; long time high intensity exercise group compared with control group, P < 0.01, there were significant differences; long time high intensity exercise group and aerobic the exercise group compared to P < 0.05, there was significant difference. SOD: general aerobic exercise group compared with the quiet control group, P < 0.05, there are significant differences;long time high intensity exercise group compared with the general aerobic exercise group,P < 0.01, there is a very significant difference.(4 ) The correlation between MDA and apoptosis was positive correlation, the correlation coefficient was 0.702, and the correlation was significant.Conclusion: (1) After a long period of intense exercise, morphological changes of skeletal muscles occurred in rats.(2) After a long period of intense exercise, the formation of free radicals in skeletal muscle tissue of rats increased, the scavenging ability decreased, and the skeletal muscle function decreased.(3) The apoptosis of skeletal muscle cells was significantly higher than that of the control group and the general aerobic exercise group.(4)There was a significant positive correlation between MDA and apoptosis.
