ObjectiveTo study the impact of different-temperature exercise on rats’ skeletal muscle apoptosis. Find the strengths and weaknesses between rats do exercise in low temperature or high temperature and rats do exercise in room temperature. Provide some references for the fitness theoretical and practical content.Methods40male six-week-old SD rats are randomly divided into four groups(n=10):normal control group,low temperature swimming group,room temperature swimming group,high temperature swimming group.Each group take exercises under prescribed water,6times a week,every30min. Four weeks later,we observed the situation of skeletal muscle cell apoptosis by HE staining,TUNEL and immunohistochemical staining.ResultsWe found a series of changes in the rats’ skeletal muscle fibers after HE staining,including muscle bundle boundaries began to blur, muscle fibers are arranged not neat,diffusion layer between the muscle fibers, increase the gap between the muscle fibers, connective tissue hyperplasia, inflammatory cell infiltration, or accompanied by edema and apoptosis,etc.Most obvious changes occurred in the high temperature sports group;After exercise, the apoptosis rate of the gastrocnemius and Caspase-3expression was significantly higher than that in the control group (P<0.05);Compared to the rats which did sports in the room temperature environment, the rats which in the low and high temperature environment increased the number of apoptotic cells while Caspase-3positive expression rate also increased (P<0.05); The most obvious changes in skeletal muscle fiber morphology, the percentage of apoptosis and Caspase-3expression have occurred in high temperature exercise group. ConclusionsExercise can induce apoptosis in skeletal muscle,and skeletal muscle cell apoptosis will increased in the dual role of abnormal temperature and motion, which is most obvious aggravating circumstances to the hyperthermia group, suggesting that movement have to speed up the update rate of the rat muscle cells,those cells can not adapt to the body, movement needs to choose the form of apoptosis in the competition to the end of life, and those cells retain robust, meet muscular physiology in rats’ needs. Movement in high or low temperature environment exacerbate the update rate, which is more obvious at high temperature.
