Aims:This paper aims to explore the hydrogen water on strength and exhaustive exercise in rat skeletal muscle oxidative stress damage,for hydrogen water as a new type of antioxidant supplements development provide experimental basis.Method:3months old SD male rats, a total of40, weights about180-220g, are randomly divided into exhaustive exercise with hydrogen rich water group, exhaustive exercise with water group, high intensity exercise with hydrogen rich water group, high intensity exercise with water group and normal control group, each8. Adopting national standard rodent drying feed diet in each group, free diet. Each group have adaptive training treadmill of3days before formal exercise, initial training speed is5m/min, training time is3min, and then lOm/min, training time is10min, after rats adapt to incline, training intensity, time and speed gradually increased to15m/min, training time is7min, exercise time was20min.3days later, exhaustive exercise with hydrogen rich water group and exhaustive exercise with water group begin high intensity treadmill training for3weeks, training speed and training time is15m/min,5min;20m/min,5min;25m/min,50min; The forth week begin exhaustive exercise, training speed and training time is15m/min,5min;20m/min,5min;25m/min,5min;30m/min,5min;35m/min, until exhaust; high intensity exercise with hydrogen rich water group and high intensity exercise with water group begain4weeks high intensity treadmill training, training speed and training time as same as exhaustive exercise with hydrogen rich water group and exhaustive exercise with water group’s high intensity treadmill training. In accordance with the training program, weekly training5d, continuous training4w. Exhaustive exercise with hydrogen rich water group and high intensity exercise with hydrogen rich water group have gastric lavage with4ml hydrogen rich water to each rat befor exercise of30min. exhaustive exercise with water group and high intensity exercise with water group have gastric lavage with4ml water to each rat befor exercise of30min. After4weeks, rats in each group are drawn24h after the end of the last movement. Rats in each group were randomly selected2abdominal perfusion fixed, and take quadriceps tissue for histological producer, electron microscopy of rat skeletal muscle ultrastructural changes; After the blood of the rest of the rats werequickly removed the skeletal muscle tissue into liquid nitrogen storage, preparation of serum, plasma in-80℃refrigerator. Determination of biochemical indicators of plasma Hb content, serum SDH and CK activity and skeletal muscle of MDA, SOD and total antioxidant capacity, and Western-Blot detection of skeletal muscle muscle autophagy-related protein expression.Results:1High intensity exercise and exhaustive exercise can lead to skeletal muscle ultrastructure in exercise-induced injury occurs, the high-intensity exercise can lead to mitochondria, dilated sarcoplasmic reticulum; Exhaustive exercise leads to a further increase in mitochondrial vacuolar degeneration, injury, sarcoplasmic reticulum dilation, mitochondrial structural damage, membrane structure damage, fracture phenomenon of sarcomere.2High intensity exercise rats hydrogen rich water supplement, skeletal muscle cell edema and myofilament fracture is reduced somewhat, mitochondrial damage mitigation, structural integrity of cell membrane is better. Exhaustive exercise rats after hydrogen rich water supplement, mitochondrial structure damage have appeared, but have reduced than exhaustive exercise with water group, muscle wire structure is complete, the integrity of the cell membrane is better. Tips, hydrogen rich water can effectively reduce the exercise intensity and exhaustive exercise on skeletal muscle injury, protection of mitochondrial function, decreased mitochondrial vacuolar degeneration appear, protect cell membranes, mitochondria and myofilament structural integrity.3Biochemical index the results show, the high intensity exercise and exhaustive exercise causes oxidative stress levels are increased, but exhaustive exercise with hydrogen rich water group and high intensity exercise with hydrogen rich water group compared with control groups:the contents of plasma Hb (p<0.05), serum CK activity (p<0.05) and the skeletal muscle MDA levels (p<0.01) was significantly lower, serum SDH, skeletal muscle SOD activity and total antioxidant capacity were significantly higher (p<0.05). Hydrogen water that can effectively reduce the intensity and exhaustive exercise in rat skeletal muscle oxidative stress levels, increase the activity of antioxidant enzymes and total antioxidant capacity.4High intensity and exhaustive exercise in rat skeletal muscle after hydrogen water, expression of mTOR protein and in comparison to the control group increased significantly (p<0.05). High intensity and exhaustive exercise in rat skeletal muscle after hydrogen water, expression of PmTOR protein and in comparison to the control group increased significantly (p<0.01).5High intensity and exhaustive exercise in rat skeletal muscle after hydrogen water, expression of LC3B-2protein and in comparison to the control group reduced significantly (p<0.05).6High intensity and exhaustive exercise in rat skeletal muscle after hydrogen water, expression of P53protein and in comparison to the control group reduced significantly (p<0.05).7Exhaustive exercise rats supplemented with hydrogen water can effectively prolong the time of exhaustion, improving sports ability.Conclusions:1High intensity exercise and exhaustive exercise can lead to skeletal muscle ultrastructure in exercise-induced injury occurs, the high-intensity exercise can lead to mitochondria, dilated sarcoplasmic reticulum; Exhaustive exercise leads to a further increase in mitochondrial vacuolar degeneration, injury, sarcoplasmic reticulum dilation, mitochondrial structural damage, membrane structure damage, fracture phenomenon of sarcomere.2Hydrogen rich water can effectively reduce the high intensity exercise and exhaustive exercise on skeletal muscle injury, protection of mitochondrial function, decreased mitochondrial vacuolar degeneration appear, protect cell membranes, mitochondria and myofilament structural integrity.3Hydrogen rich water can effectively reduce skeletal muscle oxidative stress injury of skeletal muscle, improve the antioxidant ability, improving sports ability.4High intensity and exhaustive exercise can improve the rat skeletal muscle cell autophagy phenomenon, resulting in skeletal muscle cell apoptosis. Hydrogen rich water can effectively restrain the rat skeletal muscle cells overexpression of autophagy which in intensity exercise and exhaustive exercise.
