Phenolic compounds are plant secondary metabolites of weight, it is to make plant solidpart of one of the main material. Wine in the phenolic substances it is complicated in structure,variety, a lot of people have positive effects on the body of some chronic diseases such asheart disease, cancer has certain treatment and preventive action. However, at present still didnot see wine a variety of free phenol and detection method standard to identify the winequality, this article mainly research contents are as follows:To wine gallic acid, resveratrol, quercetin and nine major free phenol, with single factorexperiment on the solid phase extraction of elution solvent, eluent volume and eluent of pHvalue and liquid-liquid extraction extraction solvent, extraction liquid product, pH value andextraction time study choice, and established a phenolic compounds in high performanceliquid chromatographic condition, the establishment of a wine phenolic compounds in optimalexperiment scheme: the liquid liquid extraction method: using ethyl acetate as extractionagent, in the pH=2condition,150ml ethyl acetate three times for extraction, extraction timefor10min. The Venusil ASB C18(5μ m,150(?),4.6x250nm) chromatographic column,mobile phase of2%acetic acid aqueous solution and acetonitrile mixed solution, usinggradient elution, flow rate is1.0mL/min, detection wavelength of280nm, columntemperature30℃. Method detection limit is low, recovery are86.295.3%between the freephenol and the relative standard deviation is less than3. And then tested the three differentred wine in the content of phenolic substances. Results show that the wine in the gallic acid,catechin, rutin and phenols content is higher, content in the range between15.0and300.0.
