Agrocybe aegerita is one of the special agricultural products in Fujian province,which contains rich nutrition and medicinal efficacy. In this paper,polyphenols wereextracted as the material from Agrocybe aegerita,the technology of extractingpolyphenols from Agrocybe aegerita with Solvent extraction,Media Optimization forextracting polyphenols from Agrocybe aegerita with supercritical CO2process byResponse Surface Method,the purification,antioxidant activity and the impact todrosophila melanogaster’s lifespan were studied. The approaches of deep processingof Agrocybe aegerita is expanded, the basis data and scientific references are offeredfor the research of Agrocybe aegerita function studies.An solvent extraction process of Agrocybe aegerita polyphenols was studied inthis paper. Different factors were considered,such as extract temperature,ethanolconcentration,extract time,material-solvent ratio. And orthogonal tests were alsodone based on the single factor tests.It was indicated that the optional conditions for organic extraction was:the watertemperature was80℃,ethanol concentration was50%,extraction time was3hours,material-solvent ratio was1:30,the extraction rate could reach to14.96%. Thestrengthen of solvent extraction process of Agrocybe aegerita polyphenols byultrasonic was further study. Different factors were considered,such as the ultrasonicpower,ultrasonic frequency,ultrasonic time,extracting times. And orthogonal testswere also done based on the single factor tests. It was indicated that the optionalconditions for ultrasonic extraction was:ultrasonic power was240W,ultrasonicfrequency was80Hz,ultrasonic time was80min,ultrasonic times was2,the extractionrate could reach to16.82%.The extraction and isolation conditions of Agrocybe aegerita polyphenols bysupercritical CO2process were studied in this experiment. Different factors wereconsidered, such as the extraction time,extraction pressure,extraction temperature,CO2Current capacity,entrainer amount. Response surface methodology was used tooptimize the extraction rate of Agrocybe aegerita polyphenols and the regression equation was established. The optional process of supercritical CO2process extractiontechnology were: extraction temperature48.49℃,extraction pressure30.62MPa,extraction time131.25min,CO2current capacity14L/h,entrainer amount90mL,theextraction rate was17.64%.Through static adsorption and desorption experiments,the purification ofAgrocybe aegerita polyphenols were researched. HPD-600was selected as the resin topurificate Agrocybe aegerita polyphenols among five macroporous adsorption resins.To research the adsorption effect and desorption effect,some factors wereconsidered,such as pH of Agrocybe aegerita polyphenols,concentration of Agrocybe aegerita polyphenols,sample volume,rate of sampleloading,ethanol concentration and elution speed. It was indicated that the optionalconditions for adsorption were:pH4.0,concentration was3.0mg/mL,sample volumewas5BV,rate of sample loading was3BV/h,the optional conditions for desorptionwere:ethanol concentration was60%,elution speed was5BV/h. polyphenol contentof purificated Agrocybe aegerita polyphenols was73.6%.The total antioxidant capacity、scavenging·OH、·O-2and·DPPH of Agrocybeaegerita polyphenols were researched, which were compared with the effect of Vcand tea polyphenol. The result showed that Agrocybe aegerita polyphenols couldeffectively scavenge·OH and·O-2,and have the scavenge of total antioxidantcapacity;Agrocybe aegerita polyphenols have lower ability on·OH compared with teapolyphenol and stronger scavenging ability on·O-2and total antioxidant capacitycompared with VC.Taking wild type Drosophila melanogaster as trial object,the influence ofAgrocybe aegerita polyphenols on the longevity of Drosophila melanogaster wasresearched,0.25mg/mL、1mg/mL、4mg/mL Agrocybe aegerita polyphenols wereadded to culturemedium of Drosophila melanogaster,observe the survival ofDrosophila melanogaster and calculate the median death time、the average lifespan、the maximum average lifespan、life extension rate. The result showed that the mediandeath time、the average lifespan、the maximum average lifespan of various dosagegroups Drosophila melanogaster are higher than control group. The Drosophila melanogaster of middle dose group of the median death time has the biggestdifference,the average lifespan、the maximum average lifespan have reachedextremely remarkable level compared with control group. The life of all dose groupshave prolonged in different extent,The rate elongation of Drosophila melanogaster ofmiddle dose group is higher than low dose group and high dose group. FemaleDrosophila melanogasters are more sensitive than male ones with Agrocybe aegeritapolyphenols.
