Margarita is plant which belongs to Rutaceae,Citrus Subfamily,Citrus Family,Margarita,and is One of the characteristics of fruit in Fujian.With the rapid development of processing industry, the lots more abandoned Margarita seeds not only caused a waste of resources ,but also cause the environmental pollution.Extraction technology and the oil property value were studied in this paper.And then the oil composition of Margarita seeds were analyzed by GC-MS.The effect of scavenging·OH and·O2- and the total antioxidant capacity were also detectionde in this paper.By the way discussion the feasibility and nutritional as edible oil. Therefore, an important recommedation for developing deep-processing products of Margarita seed oil were offered.(1)An solvent extraction process of Margarita seeds was studied in this paper. Different factors were considered, such as the solvent, extract time,material- solvent ratio,extract temperature. And orthogonal tests were also done based on the single factor tests. It was indicated that the optional conditions for organic extraction was: petroleum ether as solvent, the material- solvent ratio was 1:7, the water temperature was 60℃,extraction time 2 hours, the extraction rate could reach to 38.90%.The strengthen of margarita seeds oil solvent extraction by ultrasonic was further study. Different factors were considered, such as the power, frequency,extract time. And orthogonal tests were also done based on the single factor tests. It was indicated that the optional conditions for ultrasonic extraction was:Petroleum ether was used to extract oil from Margarita seeds by 1:7,under 150 power,80 frequency three times and 30 minutes for each time, the extraction rate could reach to 42.41%.Ultrasonic assisted extraction improved significantly extraction ratio and shortened extraction time than solvent extraction method.(2)The extraction and isolation conditions of Margarita seed oil by supercritical CO2 process were studied in this experiment. Different factors were considered, such as the extraction time,extraction pressure, extraction temperature,CO2 Current capacity,analytical pressure,analytical temperature,and suitable conditions were determined based on the single-factor and orthogonal experimental tests. The best extraction technology were:extraction temperature 40℃,extraction pressure 30MPa,analytical pressure10MPa,analytical temperature55℃,CO2 current capacity 15L/h,extraction for 2h, the extraction rate could reach to 45.1%.The extraction rate of supercritical CO2 fluid was higher 8.2% than solvent extraction,and higher 6.34% than Ultrasound-assisted extraction.(3)The acid and peroxide value of margarita seed oil extractioned by supercritical CO2 fluid were slightly lower than solvent and ultrasonic assisted extraction method. May be due to supercritical state is not conducive to fat oxidation, inhibit the formation of free fatty acids. However, saponification and iodine value were slightly higher than the other two methods, show that supercritical fluid extraction method was moreconducive to margarita oil extraction in the unsaturated fatty acids. Combination of extraction rate, can be introduced that ,supercritical CO2 extraction was more conducive to margarita seed oil extraction.The physical and chemical properties of margarita seed oil extractioned by supercritical CO2 were as follows: Saponification value of 200±3.45,explain that margarita seed oil had middle molecular weight. Iodine value was 130.12±2.67, description that margarita oil was semi, in addition to containing oleic acid, and linoleic acid, also contains a lot of double bonds. Analyzed that margarita seed oil contained a large number of unsaturated double bond, by the way has a high utilization value by its Refractive index was 1.467,which was significantly higher than normal oil. The chemical properties of margarita seed oil were keeping with Health standards of National Edible Oil by Proportion of 0.918, acid price of 1.58±0.06 and Peroxide value of 5.02±0.14.(4)Fourteen compounds of Margarita seed oil were detected by GC-MS,The main components are linoleic acid (47.82%),oleic acid (17.06%),palmitic acid (15.88%),9 (Z) - octadecadienoic en -1-- alcohol (7.21%),stearic acid (4.55% ), unsaturated fatty acid content of up to 64.88%.The linoleic acid of Margarita seed oil was generally higher than rapeseed oil, linseed oil, peanut oil and sesame oil. The content of linoleic acid was 165.6%higher than Rapeseed oil which contents 18%,and 112.5%higher than Linseed oil, which contents22.5%,show that Margarita seed oil had a value to development. (5)The total antioxidant capacity of Margarita seed oil was studied by FRAP, and effect on scavenging·OH and·O2-were studied by Fenton reaction and Pyrogallol Autoxidation Method respectively, which were compared with the effect of Vc. The result showed that Margarita seed oil could effectively scavenge·OH and·O2-,and havd the strong total antioxidant capacity. Margarita seed oil had stronger scavenging ability on·OH and lower ability on·O2-compared with Vc.
