With the increasingly serious environmental pollution, soil, air and water are subject to the influence of different levels. Many brewing processes are dependent on specific microbial communities, changes in the environment will directly alter the structure of microbial community, which in turn affect the production stability. Therefore, testing regularly the composition of microbial community is very necessary, but now the technologies that test microbial communities are slow, so we need make research on rapid quantitative technology. This subject conducts a preliminary study on this technology, mainly using the method of real-time PCR to distinguish microbial strains that have different Tm values for the same PCR samples. At last, we screened out PCR enhancers and enhancer combinations. They can partially distinguish the microbial communities.This subject makes the research in three areas, at first the brewing microbes are identified, mainly microorganisms in Jiaoni, we analyzed the main species and content of microorganisms, then compared with some standard microorganisms in the Daqu; secondly, we made QPCR experiment by using13kinds of single enhancer and78kinds of enhancer combinations to see if we can choose out of effective enhancers for a clear distinction of the main species; finally, we simulated the proportion which stands for main species’proportion in Daqu samples by using16SrDNA templates, analyzed that if this method could detect the changes of the ratio of different microorganisms, and added the PCR enhancers to the system to understand the enhancers’impact.Through the above experiments, we can get the following conclusions:the types and the content of microorganisms in Daqu sample are more than that in Jiaoni, they are different in microbial composition; screened out9kinds of single enhancer and12kinds of enhancer combinations for a clear distinction of the main species in Daqu of GuJing liquor,then by futher screening, we got single enhancer betaine and enhancer combinations betaine and ethylene glycol; used QPCR method and derivative plot method can detect changes in the proportion of mixed template.
