In order to investigate distribution of allergenic pollens in Shanghai atmosphere,main atmospheric pollutants were collected from April2010to May2010, andpollutant concentration was obtained by calculation. Flying pollen numbers of thesame period were compared with pollutant concentrations. An exposure devicesimulating adsorption of pollutants and pollen was assembled. SEM-EDS, ESR,SDS-PAGE,2-DE and MS-MS were used to study the microstructure, sensitizationallergen protein and biological activity of plane tree pollen in Shanghai.SEM results showed that there are much more particulates adhereing the pollenwhich was exposed with gas mixture for1hour and8hours, and bump appearedoutside the pollen. Pollens collected from Baoshan district and Putuo district werealso adhered much particulate. These bump was different from Ubisch bodymentioned in early literature.When Tempone-H was added, free radical signal of pure pollen express anincreasing trend till60minutes. The longer the time of exposure of pure pollen andgas mixture is, the weaker of free radical signal is. Pollen from all sampling area hasless free radical when exposed with gas mixture. On the contrary, free radical sinalchange stronger when pollen exposed with DEPs(Disel Engine Particalates).SDS-PAGE results showed that a few more obvious protein bands can berevealed:22kDa,35kDa,39kDa,71kDa. In addition, a105kDa band can be seen inAP8DA(pollen exposed with DEPs for8hours). Band color of AP8DA is muchdarker than ohers.2-DE(Two-dimensional electrophoresis) results showed that there are morepollen points in AP1A(pollen exposed with DEPs for1hours), and color is deeper.Number of protein points of AP8A is more than APB(American pure pollen beforeexposre), and color is lighter. While number of protein points of AP8A is less thanAPB. Cobined with mass spectrometry experiment result, we found that the proteinPla a1, which is the main allergenic protein in Platanus pollen, has a greatest change during the course of8hours’ exposure.
