Optimization Of Hunan Lonicera Macranthoides Volatile Oil’s Extraction Process By SFE-CO2and Composition Analysis

Our research focused on the Lonicera macranthoides DC of Xiaoshajiang Town, Longhui, Shaoyang in Hunan Province, as well as its extraction experiment by utilizing Superitical Fluid Extraction-CO2(SFE-CO2) firstly, and then established a series of parameters to optimize this experiment. Finally, the Gas Chromatography-Mass Spectrometer (GC-MS) Method were used to identify its components and determine the relative contents.1. The SFE-CO2Facility was applied to optimize the extraction pressure parameter of the Hunan Lonicera macranthoides DC powder samples. We found that under the specified pressure and temperature, the extraction products begined producing at the pressure of14MPa or above, and reach the maximum at the pressure of20MPa.2. In the temperature parameter optimizing experiment, the data showed that under the specified pressure and temperature, the extraction products begined producing at the extraction temperature of35℃or above, and reach the maximum at the temperature of40℃, above which the extraction products decreased along with the increasing temperature.3. In the SFE-CO2isolation parameter experiment, we found that the extraction product was completely isolated at the isolation pressure of13MPa or below, but intact extraction products were obtained at the isolation pressure of13MPa or above, when the extraction product was incompletely isolated. This meant extraction products of different phases could be obtained by multi-stage isolation at this condition.4. Applying real-time mass measurement on the SFE-CO2products during the whole process, we found that when the extraction time reached180min, the extraction products remained constant, which meant the extraction process was completed.5. By applying the GC-MS Method in analyzing and identify the SFE-CO2products, we used area normalization in content determination. The data suggested that there were44components in the product totally, of which17components had over1%content; the Nanocosane had the maximum content of27.6%; and the Palmitic Acid had a maximum content of10.2percent among the volatile oil.6. Utilizing the GC-MS Method to analyze the SFE-CO2products, we found that the extraction products obtained under different extraction parameters generally had identical primary components, but some differences in the quantity of its trace components, which increased along with the increment of the extraction yield.Our research established an intact, fast way to extract volatile oil from the Hunan Lonicera macranthoides DC and its components identification. Based on the experimental results, we could analyze the advantages of the Hunan Lonicera macranthoides DC from that of other production areas, which provides reference for the following research and comprehensive utilization of the Hunan Lonicera macranthoides DC.
