Gulcoraphanin is one kind of glucosinolates. Sulforaphane, the main degradat-ion products of gulcoraphanin is one kind of isothiocyanate that has the strongestanticancer activities which find in vegetables. The content of gulcoraphanin inbroccoli seed is very rich, about20-50mg/g. There are many researchers who aretrying to take gulcoraphanin extracts as raw materials, to develop health food andmedicines both domestic and foreign. However, the premise of development andutilization of gulcoraphanin is to obtain a large quantity of standard extract.This study takes the broccoli seeds as raw material, trying to build the accuratedetermination method of gulcoraphanin, and to optimize the process of extractionand separation that can use in batch preparation. The results of the study are asfollows:(1) The new detection method by HPLC of gulcoraphanin has been establish-ed.The analysis was performed on Global Chromatography SP-120-5-C18-APcolumn, Waters2487uv detector, the detecting wavelength was229nm, the mobilephase is methanol-water=1∶9(including0.1%formic acid, pH=4.0), the flowrate was1.0mL/min. In these liquid chromatographic conditions, the isolationbetween gulcoraphanin and impurity is2.54, The recoveries of gulcoraphanin were99.45%. This method is applicable for precise quantitative of gulcoraphanin(2) The preliminary extraction process of gulcoraphanin has been optimized.Broccoli seed (including gulcoraphanin18.61mg/g) that after pretreatment ofdestroy enzyme, crushing, degreasing, drying, take80%methanol water asextraction solvents, the ratio of solid to liquid is1∶20(g/mL), the temperature ofextraction is70℃, the time of extraction is20min. The extraction rate ofgulcoraphanin is92.84%.(3) The separation process of initial extract of gulcoraphanin has been optim-ized.The initial extract can be isolation by ion exchange process. D315ion-exchan-ge colophony has better adsorption effect on gulcoraphanin. The elution agent is0.5mol/L sodium chloride. When the sample volume is twenty times of ionexchange agent’s weight (g/mL), the extraction rate of gulcoraphanin in separationis89.36%. The content of gulcoraphanin in the white powder that obtained by drying andconcentrating the eluent is245.60mg/g.The total extraction rate of gulcoraphanin in exctraction and separation is82.96%...
