Astragalus root as "standard of Chinese medicinal materials in sichuan province" in2010version, is the resin for leguminous plants Astragalus ernestii Comba dry roots. It has a long medicinal history, often replaces pharmacopoeia astragalus species medicinal in sichuan area. The leading producer is in a’ba prefecture of sichuan province, ganzi prefectures, liangshan prefecture and ya ’an area and part of the mianyang county. It possess effects with supplement qi and stem desertion, disinhibit urine and draw toxin, expelpus, close sores and engender flesh.First of all, we extracted different parts from the astragalus, the petroleum ether extract and ethyl acetate extract and n-butanol extract, water-soluble part are respectively screened by mice forced swimming test on mice anti-fatigue activity, carbon clearance test in mice and hemolysin antibody generating test on the mice immune activity, we preliminarily determined the ethyl acetate extract parts as the active site.Secondly, carrying out the system preliminary experiment to test on chemical composition in ethyl acetate extract, the results showed that ethyl acetate parts may contain flavonoids, saponins composition. Combined with astragalus membranaceus modern pharmacological research results to determine flavonoids ingredients as the effective parts for anti-fatigue action. On this basis, this paper carried out the determination method research about ultraviolet spectrophotometric method of total flavonoids and HPLC of campanulin, established the evaluation method of flavonoids.Third, carry out the research about effective parts flavonoids extraction and purification technology.In the study of extraction process, we took the comprehensive scores composed by total flavonoids. campanulin and paste-forming rate with the weight coefficient of4:4:2as evaluation index, the optimization extraction technology is as follows:40times the amount of70%ethanol extract2times,1hour for each time. In the study of purification process, we researched on macroporous adsorption resin to extract the total flavonoids and campanulin purification technology, respectively on the resin type, concentration of sample solution, sample weight, the sample flow rate, elution solvent and its dosage, the final purification process conditions were determined by X-5macroporous adsorption resin, on sample liquid concentration is4.05mg/ml of total flavonoid, campanulin of0.061mg/ml, the sample amount to30ml, the sample flow rate of1.5ml/min,3BV with distilled water,50%ethanol6BV respectively according to1.5ml/min flow rate of elution, abandoned to distilled water elution part, collected50%ethanol elution part. After macroporous resin purification, purification effect of the total flavonoids and campanulin is more than3times,purification effect is good.In conclusion, we determined the ethyl acetate extract parts of astragalus as effective parts for anti-fatigue activity of mice, the immune activity of, set up the the total flavonoids and campanulin determination method, following the method as evaluating indicator, we preliminarily finished extraction and purification technology of the total flavonoids of astragalus. The study had a certain reference value on astragalus flavone compound preparation, quality standard,pharmacokinetic and pharmacodynamic study, laid a foundation for the quality standard of astragalus total flavonoid parts research and development and utilization,provided pharmacological basis for the clinical standard medication.
