Extracellular fatty acid binding protein (Ex-FABP) is a member of the FABPs and one of the impoetmant regulatory factors in the body’s fat metabolism process. It is believed to participate in the regulation process of fatty acids, muscle fibers and skeletal in chicken. Adducin is one of the cytoskeletal proteins. It is thought to regulate singal transduction, cell-to-cell contant, and cell membrane ion transport to help fat metabolism. This study have scanned the genetics diversity of Ex-FABP and ADD1genes among Fast Partridge, D-Dwarf chickens, Guangxi local breed2chickens, Silkies and lingshan chickens using molecular biology techniques, and analyzed the association between the polymorphisms and growth and adipose traits; simultaneously detected their expression in various organizations by real-time fluorescence quantitative PCR. The main results were as follows:1. Effects of polymorphisms of Ex-FABP gene on carcass and adipose traits of chickensWe have detected three genotypes(AA,BB,AB) of Ex-FABP gene by SB-ASA in chickens. The result of different genotypes and chicken’s carcass and adipose traits showed that:There was no significant difference in all traits in Fast Partridges and lingshan chickens; AB genotypes had significant enhance effect on the abdominal fat percentage (AFP) than AA genotype in female D-Dwarf chickens; BB genotype was more superiority than AA and AB on the abdominal fat (AF) and AFP in male Guangxi local breed2; BB genotype had significant higher AFP than AA and AB in female Soil two chickens; in male Silkies, BB genotype was better than AB on the leg percentage(LP).2. The relationship between polymorphisms of ADD1gene and carcass and adipose traits of chickensBy PCR-RFLP and DNA sequencing, the polymorphism of ADD1gene on exon8was studied. The mutation of ADD1gene did not lead to a substitution of the amino aeid and three different genotypes (TT,CC,TC)were found by SB-ASA. The associations analysis between the genotypes and carcass and adipose traits revesled that:the homozygous(TC) birds had significant higher LP than TT and CC in female Fast Partridges; TC genotype was bigger than CC in weight of breast muscle in male D-Dwarf chickens; TC genotype had significant influence in eviscerate percentage(EP) than CC, and was better than TT in breast weight in male Guangxi local breed2; leg weight(LW) and leg percentage(LP) in genotype TT were bigger than the other genotypes in female Guangxi local breed2; in Silkies, different genotypes didn’t significantly influence carcass and adipose traits; TC genotype had significant enhance effect on the body weight(OW), caress weight(CW), halfeviscerate weight(HW) than CC, weight of breast muscle in genotype TT and TC was bigger than CC, LP content of the TT genotype was significantly higher than CC genotype.3. Expression profile of Ex-FABP and ADD1genes in chickenExpression pattern of Ex-FABP and ADD1genes in the thirteen different tissues of chickens were studied by real-time fluorescent quantitative PCR. The results showed that:for Ex-FABP gene, the expression of lung was the highest, and then crrebellum, pituitary and abdominal fat, heart, liver and leg muscle were low, of spleen, kidney, pancreas, brain, hypothalamus and skeletal muscle were the lowest. For ADD1gene, the highest expression level were detected in heart, birin, hypothalamus and pituitary, then in kidney and leg muscle, less in liver, spleen, lung, pancreas and abdominal fat, and lowest in cerebellum and skeletal muscle.
