Intramuscular Fat(IMF) has important effect on meat sapor and delicate flavour,and has become the important index of meat quality scaling. IMF is deemed to bepositive correlation with meat palatability, and it may affect meat sapor, succulence andtenderness. Presently, the improvement in poultry breeding has been made. However,negative effects have appeared accompanied with high intensity selection. Problems neededresolved by poultry industry in China, exist in controlling excessive fatness accumulationthat affects chicken carcass quality. This study was designed to investigate the effect ofH-FABP gene on chicken meat quality, at the same time, this study is conducted tounderstand the genetic variation and the genotype diversity, then serve this gene as geneticmarker and use in maker assisted selection, and establish the element of breeding highquality chicken.Using PCR-SSCP technology, this study designed to investigate the effect of H-FABPgene on chicken fat metabolize in four population of 4 parochial breed of Fengkai Xinghuachicken etc and 2 lingnanhuang crossbreeds and filtrate 4 loci that have distinct correlationwith Ventral Fat Rate(VFR) and Intramuscular Fat(IMF), then take the 4 loci as assistedmaker of VFR and IMF selection by SNPs detection. We detected the genotype of the 4 lociin generation 1 of a new yellow broiler line and got genotypes on 4 loci. Then based ongenotypes of each individual at 4 SNP sites, predicted breeding values as selection indexwere applied into the selection scheme. Generation 1 of the population was divided into coregroup, high fat group and low fat group according to the selection index or normalphenotype selection. Generation 2 were reproduced in each group of selection. We detectedthe genotype of the 4 loci in generation 2 by SNPs detection and analyze the effect of geneassisted selection.Results indicate that the frequency of gene and genotype on the 4 loci betweengeneration 1 and generation 2 changed. On site 1, the frequency of allele A has changedfrom 0.5727 in generation 1 to 0.3037 in generation 2. On site 2, the frequency of allele A has changed from 0.5396 in generation 1 to 0.5870 in generation 2. On site 3, thefrequency of allele A has changed from 0.5718 in generation 1 to 0.4145 in generation 2. Onsite 4, the frequency of allele A has changed from 0.5617 in generation 1 to 0.4468 ingeneration 2. We analyze the difference of Ventral Fat Rate (VFR) and Intramuscularfat(IMF) among the core group, high fat group and low fat group and found the differenceamong those 3 groups is not remarkable on those two traits. The VFR of the core group andthe high fat group was trending to be higher. Results indicate the maker assisted selectionusing pro- gained loci which have distinct correlation with fat quality not arrive at ananticipative selection popurse.Base on the determinant on genotype and VFR and IMF of every individual, thisStudy has analyzed the character significance of different genotype on every locus. As aresult, genotype has no significant effect on IMF and VFR (P>0.05). But in the highfat-population, the least square means of IMF of genotype AA is obviously higher thangenotype BB and AB (P<0.05) on locus 2. In the rambreeding-population, the least squaremeans of IMF of genotype AB is obviously higher than genotype BB and AA (P<0.05) onlocus4. United the 4 loci as one gene, we analyzed the character difference of differentgenotypes combination, as a result, the genotypes combination had no significant effecton VFR and IMF. Results indicate that there were no significant difference betweengenotype in other makers on VFR and IMF.Results indicate the correlation between specifically gene variation and quantity traitsmay be distinct in different population and different hereditary background. Themaker-assisted selection on our chicken population using the loci which filtrating fromFengkai Xinghua chicken etc not arrive at an anticipative selection popurse. Besides, themakers that have significant in other breeds have no significant with FVR and IMF in thispopulation. The 4 loci we studied on the H-FABP gene is not the fittest candidate marker asthe Marker-assisted selection on IMF and VFR in this line of yellow broiler. Moremolecular makers really correlated to VFR and IMF must be filtrated in this population ifwe want to make use of the means of marker-assisted selection in seed selection.
