The frozen semen of Holstein bulls were identificated in order to establish a simple and practical technology for bull individual identification.The relationships between polymorphisms of the BMP7and USP26gene and the sperm quality traits were analyzed in Chinese Holstein bulls, in order to select molecular makers for improving the breeding efficiency of bulls.I.STR was amplified with fluorescence labeled primers of17microsatellites, Genetic Analyzer (AB3130x1) was used to separate and analyze the genotype of the amplification products. The individual identification of bulls were identified in this experiment with the standard that only one locus mismatch was permitted.The results showed that all the17microsatellite loci had high genetic polymorphisms and the Cumulate DP (CDP) was99.99%. the cumulate discrimination power was99.99%.This study suggest that the17microsatellites loci of this experiment could be used in the bull individual identification.II.By PCR-RFLP and sequencing, the single nucleotide polymorphisms of two locus in BMP7gene intro5and USP26gene exon in171Holstein bulls were analyzed.The associations between the polymorphisms of the genes and the traits of the bulls’frozen semen were analyzed.The results showed that at locus BMP7gene A76045G, the bulls’ frozen semen motility with genotype GG was signifycantly higher than those with genotype AA(P<0.05); At locus USP26gene C1156T, the bulls’sperm density with genotype CT and CC were significantly higher than those with genotype TT(P<0.05).The two SNPs had no significantly relationship with ejaculate volume and frozen semen motility(P>0.05).BMP7gene and USP26gene may be regard as a choice of molecular makers of Chinese Holstein bulls with high sperm quality in the future.
