Hericium erinaceous polysaccharide is the main active substances. At present, most of Hericium erinaceus products in the market are produced by simple processing, which contains many impurities. Such as Hericium erinaceus polysaccharide products, which are crude polysaccharide without optimized separation, contain more protein, pigment, and the color is darker and can not perform the best effect of polysaccharide. So the research on preparation process technology used in industrial production of crude polysaccharide and how to obtain polysaccharide with high biological activity is necessary. This paper mainly studied on the water extraction and alcohol preparation technology parameters optimization of Hericium erinaceus crude polysaccharide, Hericium erinaceus polysaccharides decoloring method and the chemical properties and biological activity after decolorization. This results could be used as theoretical foundation for Hericium erinaceus polysaccharide depth development. The main content of the paper are as follows:1. The factors which affect the polysaccharide yield and content during the water extraction and alcohol precipitation of the Hericium erinaceus polysaccharide preparation were studied. And drying methods for the polysaccharide are also researched. Then through orthogonal test, the optimized factors were selected, the optimize conditions are conclude. Extraction in hot water two times, final ethanol concentration is70%, liquid material ratio is15:1, and enrichment degree is3:1. By the extraction of several kinds of enzyme contrast with reference method, it is found out that most of the single enzyme can not improve yield and content of the polysaccharide.2. For higher decoloring rate, polysaccharide retention rate and immunity activity in vitro, three commonly decoloring methods are compared, and the conclusion is that the macroporous resin is more suitable for the decolorization of Hericium erinaceus polysaccharide. Ten types of macroporous resin are studied and the results show that macroporous alkalescence anion resin D315is the most suitable resin for decolorization of Hericium erinaceus polysaccharide. For higher decoloring rate, polysaccharide retention rate, several factors are investigated. Apply to the Box-Behnken experimental design principle based on the single factor experiment, response surface analysis method is designed with three factors and three level, and two regression model on the decoloring rate, polysaccharide retention rate and resin volume (mL), decoloring time (h), sample concentration (mg/mL) are established. Based on the analysis of the isopleth diagram showing in the response surface graph and the analysis of regression model optimized mathematical, the optimized decoloring conditions are conclude:100mol polysaccharide solution join resin37.12mL, decoloring time6hours, crude polysaccharide concentration is13.85mg/mL, decoloring ratio (%) and polysaccharide retention rate (%) of the theoretical value is70.59%and81.12%, respectively. On the basis of the static decoloring, dynamic decoloring technology are obtained: velocity of0.6BV/h, sample weight on270mL. After simple pretreatment, the macroporous resin D315can be used above eight times.3. Physical and chemical properties of Hericium erinaceus polysaccharide before and after decoloring(polysaccharide, protein content, molecular weight distribution, simple sugars, characteristics of groups), immunity activity in vitro and function on rat gastric mucosa damage repairing are compared. Results shows that after Hericium erinaceus polysaccharide decoloring, polysaccharide content improve slightly, protein content descend significantly and the monosaccharide proportion declined slightly: galactose and glucose ratio increased slightly, C=O group vibration decrease. β configuration polysaccharide is contained, which could contribute to the decoloring process enrichment.The immunity activity in vitro of Hericium erinaceus polysaccharide after decoloring dramatically improved (p<0.05). The function of gastric mucosa damage repairing is enhanced, which shows that Hericium erinaceus crude polysaccharide pigment and protein may have weaker or none immune activity in vitro. After decoloring process, this part of the pigment is adsorbed. The active polysaccharides get enriched and show higher activity. But whether the resin contains toxic compounds is remain to be studied in the future.
