This paper mainly studied about the cultivation of Hericium erinaceus by JUNCAO technology and Screening the formula of culture medium and the cultivation management process; furthermore, the chemical compositions of Hericium erinaceus fruit body by different formula of culture medium and the same formula of culture medium between different growth period were compared and analyzed; At the same time, a preliminary study of extraction, separation and purification of Hericium erinaceus polysaccharide was been done in this paper. The main research results are as follows:1. Using level 3 system methods to screen the most suitable formula of JunCao culture medium. Which was Dicranopteris dichotoma:39%, Neyraudia:39%, Gypsum:2%, Bran: 20%, Moisture content:60%.2. The chemical compositions of Hericium erinaceus fruit body is diversity by different formula of culture medium and the same formula of culture medium between different growth period.Specific results are as follows:In five kinds of culture medium formula, Hericium erinaceus fruit body’s crude protein content under JunCao cultivation is the highest.It is about 1.14times than traditional sawdust cultivation.The crude protein content by the same formula of culture medium is rising from mushroom budding period to Maturation period while Declining obviously in Senescence period. Moisture is the highest in mushroom budding period,but the general trend is down during the whole Growth period. Ash, crude fat and total sugar content is rising with fruit body growth and reaches a high in aging pHase.3. Using L9 (34) Orthogonal design experiments to optimize extract conditions of Hericium erinaceus fruit body’s polysaccharides. The results show that the best condition is: extract 2 times, ratio of material to water 1:15, time of 4 hours and temperature of 100℃.4.Using DEAE SepHarose Fast Flow column chromatograpHy to separate and purificate preliminarily Hericium erinaceus Fruit body polysaccharide under JUN-CAO cultivation and traditional sawdust cultivation. The results show that Hericium erinaceus Fruit body polysaccharide under JUNCAO cultivation has three components,there are cHEP1,cHEP2,cHEP3;The polysaccharide under traditional sawdust cultivation has only two components,there are sHEP1,sHEP2.Using SepHadex G-100 column chromatograpHy to identificate various components’ purity.The experimental results show that the peaks of cHEP2, cHEP3, and sHEP2 are all single symmetrical peak.
